Abstract:Cholangiocarcinoma is the intrahepatic or extrahepatic bile duct epithelial tumor with a high death rate. Cholangiocarcinoma is not sensitive to radiotherapy and chemotherapy. Hypoxic microenvironment is one of the inherent features for most solid tumors. This is accounted for by the mechanism of neoangiogenesis, by which vessels around tumor are overgrowing; vasculature distribution is in mess; and tumor vascular tissue structure is abnormal. Therefore,factors induced by hypoxia becomes one of the specific targets in cancer therapy.Objective:To investigate the expression of c-MET and HIF-1α proteins in cholangiocarcinoma tissues and analyse their association with invasion and metastasis.Method:Collected62cases of cholangiocarcinoma tissues from patients operated in Hepatobiliary and Enteric Surgery Research Center and kept in Department of Pathology, and collected15cases of normal Bile duct tissues. Using immunohistochemical staining (SP method) to detect the expression of c-Met and HIF-la in these tissues. Data were analyzed using SPSS19.0statistical softwareResults:①In cholangiocarcinoma, c-Met and HIF-1α were significantly expressed (P<0.05). The positive rates of c-Met and HIF-1α expressions were69.35%(43/62) and77.42%(48/62), respectively. Whereas, the positive rates of c-Met and HIF-la expressions in normal bile duct tissues were only6.67%(1/15) and6.67%(1/15), respectively. In other normal tissues there were negative expression.②Both carcinogenic factors were also significantly expressed under the conditions of cancer cell differentiation, pathologic progression, lymph node metastasis (P<0.05).. Gender, age and tumor size did not play a role in association with the level of c-Met and HIF-1α expression.③c-Met expression had significantly positive correlation with HIF-1α expression (P<0.05).Conclusion:The overexpressions of c-Met and HIF-la have significant association with cholangiocarcinoma development and metastasis.
