Objective: Comparing the differences of expression levels of miRNA-16,miRNA-150 and miRNA-155 in peripheral blood mononuclear cells(PBMCs)of patients with rheumatoid arthritis(RA),high-risk RA populations and healthy people.Analysing of patients' miRNA-16,miRNA-150 and miRNA-155 expression levels with their own anti-cyclic citrullinated peptide antibody(CCP),rheumatoid factor(RF),C-reactive protein(CRP),erythrocyte sedimentation rate(ESR)to find whether there is a correlation of the results of preliminary exploration of miRNA-16,miRNA-150 and miRNA-155 in the pathogenesis of RA.Providing a reliable laboratory markers for the diagnosis of RA.Methods: Seventy-one patients were selected according to the diagnostic criteria of rheumatoid arthritis: 46 patients with high-risk group of rheumatoid arthritis(EULAR score less than 6),and 40 healthy as control group,taking 2 ml of fasting peripheral blood.In order to extraction of total RNA form peripheral blood mononuclear cells,the method of differential density gradient centrifugation isolated was also used.The levels of miRNA-16,miRNA-150 and miRNA-155 was detected with the technique of the real-time PCR from the cDNA which was obtained from total RNA which was reverse transcribed.Compared with the results of routine clinical diagnosis of rheumatoid arthritis,CCP,RF,CRP and ESR,the relationship between miRNA-16,miRNA-150 and miRNA-155 and rheumatoid arthritis was further analyzed.A receiver operating characteristic curve(ROC curve)was constructed to analyze the value of combined detection of miRNA-16,miRNA-150 and miRNA-155 and the three to diagnose rheumatoid arthritis by the area under the curve(AUC).Results: 1.There was a positive correlation between the expression of miRNA-16 in peripheral blood mononuclear cells of patients with RA and its CCP,RF,CRP and ESR(p<0.05).The expression of miRNA-150 was also related to its CCP,RF,CRP and ESR in a positive correlation(p<0.05).But there was a positive correlation between the expression of miRNA-155 and the results of CCP,RF,and CRP(p<0.05),and there was no correlation between the results of ESR and the results of ESR(p >0.05).2.There was a positive correlation between the expression of miRNA-16 in peripheral blood mononuclear cells and the results of CCP,RF(p<0.05).There was no correlation with the results of CRP and ESR in the high-risk group(p>0.05).There was a positive correlation between the expression of miRNA-150 and its CCP(p<0.05).There was no correlation with the results of RF,CRP,and ESR(p>0.05).There was no correlation between expression of miRNA-155 with the results of CCP,RF,CRP and ESR(p>0.05).3.The expression of miRNA-16 in peripheral blood mononuclear cells can distinguish between RA patients and healthy control group,high-risk group and healthy control group(p<0.05),but it can not distinguish between RA patients and high-risk groups(p> 0.05);miRNA-150 expression levels can distinguish between RA patients and healthy controls,and high-risk groups and healthy controls(p<0.05),but can not distinguish between RA patients and high-risk groups(p>0.05);The expression level of miRNA-155 can distinguish between RA group and high-risk group,RA group and healthy control group(p<0.05),but can not distinguish between high-risk group and healthy control group(p>0.05).4.In the building of EULAR score,on the basis of the ROC curve,the miRNA-16,the miRNA-150 and the miRNA-155 and combined,when applied to the diagnosis of RA,AUC were 0.662,0.650,0.544 and 0.814,the sensitivity was 67.6%,70.4%,64.3% and 82.9%,specificity of 54.5%,51.7%,53.8% and 65.5%.Conclusion: 1.This study found that miRNA-16,miRNA-150 and miRNA-155 became more highly expressed in PBMCs of patients with RA and high-risk RA,suggesting that miRNA-16,miRNA-150 and miRNA-155 may be involved in the pathogenesis of RA.2.There is a positive correlation between miRNA-16,miRNA-150 and miRNA-155 and CCP,RF,CRP and ESR in RA patients,suggesting that the three may be related to the severity and severity of RA.3.The expression of miRNA-16,miRNA-150 and miRNA-155 can distinguish patients with RA,high-risk groups and healthy people to some extent.4.On the basis of EULAR score,the combined use of miRNA-16,miRNA-150 and miRNA-155 in the diagnosis of RA has better diagnostic value than the single use of the three.
