Objectives: Research the effects of miR-711in gastric cancer SGC-7901cells,explore the mechanisms of cell cycle regulation and provide a new theoretical basis forthe prevention and treatment of gastric cancer.Methods: SGC-7901cells were cultured in vitro, the miR-711overexpression vector(miR-711-mimics), miR-711inhibited expression vector (miR-711-inhibitors) andempty vector (NC) transfected into SGC-7901, qRT-PCR method validation groups inmiR-711expression after transfection by MTT, flow cytometry analysis of theexperiment on SGC-7901cell cycle-related, and further explore its target genespredicted by experiment and Western Blots mechanisms.Results:72hours after transfection by fluorescence microscope counting,miR-711-mimics, miR-711-inhibitors and NC group transfected SGC-7901, measuredtransfection efficiency of more than75%; qRT-PCR detection of the experimentalgroup the relative expression of miR-711, miR-711-mimics group, miR-711-inhibitorsand expression of miR-711in the NC group were168.31times of the control group,0.269-fold,0.998-fold; MTT results show SGC-7901cells were transfected withmiR-711overexpression vector, inhibition of cell proliferation and increased withincubation time (P <0.05); in contrast, transfection of miR-711inhibited expressionvector SGC-7901cells, the cell proliferation inhibition decreased with incubation time;it shows no significant difference (P>0.05) NC group transfected with empty; flowcytometry showes that overexpression of miR-711group compared with the other threegroups, SGC-7901cells was significantly inhibition in the G1phase (P <0.05);detected miR-711-mimics group gray value CyclinD1,CDK4by Western Blotcomparison with the rest of the group was significantly higher (P <0.05), however,miR-711-inhibitors group of gray values significantly reduced, which was the lowest. Conclusions: MiR-711overexpression can block human gastric cancer SGC-7901cellproliferation by inhibiting the expression of CyclinD1/CDK4and make cell cycle arrestin G1phase, plays an inhibitory the role of genes in cancer.
