Objective:Acupuncture on intestinal calcium absorption in ovariectomized rats as a cut point of impact, observed electricity for intestinal VDR、CaBP-D9KmRNA and protein expression in ovariectomized rat model. Detecting bone density after three months, it is determined whether a successful modeling. After a successful modeling, removed the10rats in each group, removaled of the eye, taken blood and killed them. Taken bone specimens and stored. The other10rats of sham-operated group was set to the control group, Another30rats of operation group were divided into model group, electroacupuncture group and drug group, according to the random number table,10rats in each group.Acupuncture group:select two points①Guan Yuan,Sanyinjiao (double);②shenshu, after three years (double). Daily take one set point, two sets of points alternately acupuncture, electrical stimulation with Han electro-acupuncture device, continuous pulse wave, frequency3-4Hz, strength4-5mA,With local mild degree of jitter in the rat,every20minutes, once a day.Estrogen group:subcutaneous estradiol benzoate, O.lmg/kg body weight of each rat each time, once a week. Control group and model group are not any treatment.Bone density testing in rats after12weeks of treatment, and killed them. Take the small intestinal mucosa, Observed the effects of acupuncture on CaBP、VDR mRNA and protein expression in intestinal mucosa of ovariectomized rats by using the PCR method and Western blot; Take the left distal femur metaphyseal bone tissue about1cm, fixed-embedded and made no decalcification slices, after grinding the bone specimens, observed by laser scanning confocal microscope,application Bioquant98image software to measurement and analysis the static and dynamic index of bone metrology; Removal of the eye to take blood samples, determination of calcium regulating hormone levels in rats, including estradiol (E2), parathyroid hormone (PTH), calcitonin (CT), active vitamin D (1,25(OH)2D3).Results:1.Bone density testing:EA group and drug group lumbar spine BMD were significantly improved; bone mineral density of the femur have varying degrees of improvement, particularly in the drug group improved significantly.2. CaBP, VDRmRNA and protein expression:Model group intestinal mucosa CaBP, VDRmRNA and protein expression was significantly reduced compared with the control group. Drug group and EA group CaBP, VDRmRNA and protein expression compared with the model group were significantly increased.Conclusion:1. Electro acupuncture can improve lumbar spine bone mineral density in osteoporosis rats.2. Electro acupuncture can enhance the expression of VDR and CaBP-D9K in the intestinal mucosa of rats with osteoporosis in postmenopausal.3. The mechanism of electro acupuncture may be to improve gastrointestinal bioavailability of nutrients such as calcium, correct calcium malabsorption achieved.
