Objective: The purpose of the study was to investigate the effects ofLanzhusicaohuazhuojiedu Decoction on ulcerative colitis model rats throughobservating the general condition, disease activity index(DAI) scores andcontent of serum tumor necrosis factor-α(TNF-α), interleukin-8(IL-8) andinterleukin-10, and then, try to further explore the mechanism of theLanzhusicaohuazhuojiedu Decoction.Methods:60eight-week-old male Wistar rats were randomly dividedinto six groups,each group10rats.They were respectively marked for thenormal group(NG), model group(MG), high dose group (HDG), Moderatedose group (MDG), low dose group of LanzhusicaoHuazhuoJiedu Decoction(LDG) and the SASP group (SASP). Except the NG group, the rest groupswere controlled in order to make UC animal models bytrini-trobenzenesul-fonic acid/ethanol(TNBS/ethanol). After4days model-ing, the NG group continued to feed, the MG were given normal saline, theother treatment groups received liquid gavage. After2weeks continuoustreatment, observate the rats activity, body weight, stool consistency, thegeneral condition and the colonic mucosa general concept andhistopathological changes. Detected the content of serum TNF-α, IL-8,IL-10by radioimmunoassay and analyse effects of LanzhusicaohuazhuojieduDecoction on the content of TNF-α,IL-8and IL-10.Results:1The general state and DAI scoresCompared with the NG group, the general condition of rats becamelacking of energy, activity decreased, reaction reduced, eating less, weight loss,back arched, hair loose, yellowish dull fur, diarrhea or bloody mucus. After theexperiments finished, the general condition of all rats in each group had different levels of improvements, in which the recovery of the MG group wasthe worst. General condition of HDG rats were close to the NG group. Whilethe condition of MDG and SASP were slightly lower than the NG group, andLDG group were better than MG group but less than MDG group and SASPgroup, there were no significant difference between the MDG and SASPgroup.The DAI scores:Compared with the MG group(2.50±0.31),the HDG(0.37±0.35),the MDG(0.80±0.28),the LDG(1.44±0.33)and the SASPgroups (1.57±0.39)statistically significant difference (P<0.05); Compared withthe SASP, the HDG and the MDG,statistically significant difference (P<0.05);The HDG compared the MDG,statistically significant difference (P<0.05);The LDG compared the SASP,no statistically significant difference (P>0.05).2The general observation of colonic mucosa and the histopathologicalconceptThe MG group was the worst rat,s colon mucosa, while the HDG groupwas much better, closely to the NG group. The MDG group was slightly worsethan the HDG group.The SASP and The LDG groups were between the MDGand the MG groups, which has no significant difference between the twogroups.The histopathologic aspects: the NG group was the normal rat colonmucosa, while the MG group rats were colon mucosa layer missing, glandularstructures change, congestion and edema, erosion and ulcer formation was themost serious. The condition of the HDG rats colon mucosa were close to theNG group, and the MDG rats colon mucosa recovered well but slightly worsethan the HDG.The SASP group and the LDG group were between the MDGand the MG groups.3The test results of serum TNF-α,IL-8and IL-103.1The test results of serum TNF-α in each groupThe levels of serum TNF-α in each treatment group were compared withthe MG(1.30±0.12), and there was statistically significant difference betweenthem(P<0.05). The HDG group (0.65±0.11) compared with the MDG (0.82±0.10), LDG(1.02±0.20)and SASP (1.14±0.13)groups, Significantdifference can still be observed (P<0.05).Compared with the HDG group theNG group (0.55±0.11), There was no statistically significant difference(P>0.05). The MDG group compared with the LDG and the SASP, statisticallysignificant difference (P<0.05). The LDG group compared with the SASP, nostatistically significant difference(P>0.05).3.2The test results of serum IL-8in each groupThe levels of serum IL-8in each treatment group compared with theMG(0.45±0.06), statistically significant (P<0.05). The HDG(0.27±0.06)compared with the MDG (0.33±0.05), the LDG(0.38±0.07)and the SASP(0.39±0.04),statistically significant difference(P<0.05). Compared with theMDG group,the LDG and the SASP, There was statistically significantdifference between them(P<0.05). The LDG compared with The SASP, nostatistically significant difference (P>0.05).3.3The test results of serum IL-10in each groupsThe levels of serum IL-10in each treatment group compared with theMG(12.80±2.57), statistically significant different(P<0.05). TheHDG(24.69±4.05)compared with the MDG (19.05±4.69), the LDG(17.26±2.00) and the SASP (20.81±3.28), statistically significant difference(P<0.05). The SASP compared with the MDG and the LDG groups, nostatistically significant difference (P>0.05). The LDG Compared with theMDG, no statistically significant difference (P>0.05).Conclusion:1Lanzhusicaohuazhuojiedu Decoction not only can improve the UC ratsdiarrhea, mucous pus and other symptoms,but also well alleviate thepathological damages of the rats model, and showing a certain amount-effectrelationship between efficacy and medicine dose.2The mechanism of Lanzhusicaohuazhuojiedu Decoction on the trea-tment of UC may be by reducing TNF-α, IL-8and other proinflammatorycytokines, while increasing anti-inflammatory cytokines such an IL-10, whichachieve balance cytokines to play an anti-Inflammatory and regulate immune function.
