Objectives: Ulcerative Colitis is a chronic non-specific intestinalinflammatory disease, of which the pathogenesis is not known completely yet.Traditional Chinese Medicine has abundant experience to cure the disease andthe therapeutic effect is definite.LanZhuSiCaoHuaZhuoJieDu Decoction(LZSCD) is a experiential decoction of my professor. Though the observationof the general situation,DAI scores and the expression of IL-1β,IL-4andIL-10,we tried to confirm the definite therapeutic effect of LZSCD andexplore the mechanism in order to research effective drugs.Methods:60healthy Wistar rats of clean grade were randomly dividedinto six groups with10rats in each group: the normal group (NG), the modelgroup (MG), the sulfasalazine group (SASPG), the high-dose LZSCD (HDG),the middle-dose LZSCD (MDG) and the low-dose LZSCD (LDG). UlcerativeColitis animal models were making by the way of TNBS/ethanol combinativemethod. On the3rd day,after the success of modeling, NG were fed normallyand MG were given normal saline by garage, while the other group were givencorresponding drugs. In the following14days, we observed the changes ofactivity, body weight and general changes in stool. The disease activity index(DAI) score was also finished. Then rats were killed and tissue materials wereobtained. According the instructions of kits, IL-1β,IL-4and IL-10weredetected. Then we could analyze the effect of LZSCD on the ulcerative colitismodel rats.Results:1The general condition and DAI scoresCompared with NG, all those mental status, activity condition, hairglossiness, appetite, defecate character in other groups changed different degree. After the drug intervention for14days, the general condition of HDGwas worse than that of NG and better than that of any other treatment group:the activity condition, hair glossiness, appetite and defecate character almostreturned to normal. The general conditions of MDG, LDG, and SASPG werebetter than that of MG but worse than that of HDG; MDG was better thanLDG and SASPG. For the DAI score: there was statistically significantdifference between NG and the other group(P<0.05); there was statisticallysignificant difference between HG and the other treatment groups(P<0.05);there was statistically significant difference between LDG and SASPG, butthere was statistically significant difference between those two groups andMDG(P<0.05). According to the average value, it could be found that HD、MD、LD and SASP all did something good for the improvement in defecatecharacter and weight; HD did better than MD、LD、SASP;MD did better thanLD and SASP;there was statistically significant difference between LD andSASP.2The general observation and microscope observation of colonic mucosaThe general observation: compared with NG, MG was the worst whileHDG was second to NG; SASPG, MDG and LDG were worse than HDG butbetter than MG; MDG was a little better than LDG and SASPG. Microscopeobservation: compared with NG, MG was the worst while HDG was second toNG; SASPG, MDG and LDG were worse than HDG but better than MG; therewas no obvious difference among SASPG, MDG and LDG.3The content of IL-1β、IL-4and IL-10in the serum of ratIL-1β: there was statistically significant difference between MG and theother group(P<0.05); there was statistically significant difference betweenHG and the other treatment groups(P<0.05); there was not statisticallysignificant difference among MDG、LDG and SASPG(P>0.05). Accordingto the average value, it could be found that HD、MD、LD and SASP alldecreased the level of IL-1β; HD did better than MD、LD、SASP;there wasnot statistically significant difference among MDG、LDG and SASPG(P>0.05). IL-4:there was statistically significant difference between MG and theother group(P<0.05); there was statistically significant difference betweenHG and the other treatment groups(P<0.05); there was not statisticallysignificant difference among MDG、LDG and SASPG(P>0.05). Accordingto the average value, it could be found that HD、MD、LD and SASP allelevated the level of IL-4; HD did better than MD、LD、SASP;there was notstatistically significant difference among MDG、LDG and SASPG(P>0.05).IL-10:IL-4:there was statistically significant difference between MG andthe other group(P<0.05); there was statistically significant differencebetween HG and the other treatment groups(P<0.05); there was notstatistically significant difference among MDG、LDG and SASPG(P>0.05).According to the average value, it could be found that HD、MD、LD and SASPall elevated the level of IL-10; HD did better than MD、LD、SASP;there wasnot statistically significant difference among MDG、LDG and SASPG(P>0.05).Conclusion:1The experiment showed that LZSCD could significantly improve thegeneral condition of rat and the pathology of colon, and there was dose-effectrelationship: the large dose did significantly better than middle dose; themiddle dose did a little better than low dose.2Mechanism of LZSCD on UC might be adjusting the body’s immunitythrough decreasing the level of IL-1βand elevating the level of IL-4and IL-10in the serum of rat.
