| Objectives: This experiment through the establishment of UlcerativeColitis (UC) rat model to investigate effect of LanYinFengYuHuaZhuoJieDuDecoction on UC model rats.Though observing the general situation,diseaseactivity index(DAI) scores and the content of serum tumor necrosis factor-α(TNF-α), interleukin-4(IL-4) and SOD to explore the mechanism of UC andconfirm the curative effect of LanYinFengYuHuaZhuoJieDu Decoction. Inorder to provide an experimental basis for the study of disease mechanism andexplore the effective drugs.Methods:60eight-week-old clean healthy male wistar rats wererandomly divided into6groups, each group10rats,they were respectively thenormal group (N), the model group (M), the SASP group (SASP), thehigh-dose LYFYD (HDG), the middle-dose LYFYD (MDG) and the low-doseLYFYD (LDG). In addition to the N group,the remaining five groups usingtrini-trobenzenesul-fonic acid/ethanol(TNBS/ethanol) to induce UC animalmodel.On the4rd day after the success of MK,HDG,MDG,LDG and SASPwere respectively given drug lavage therapy,M was given saline lavage.After2weeks continuous treatment,we observe the mental state, activity, bodyweight,defecate character such as general situation daily.All rats weresacrificed after14days.Then we observed the colonic mucosa in general viewand histopathological changes and according to the kit detected the content ofserum TNF-α,IL-4,SOD by radioimmunoassay and analyse effects ofLanYinFengYuHuaZhuoJieDu Decoction on the content of TNF-α,IL-4andSOD.Results:1The general case and DAI scores Compared with N, Model group rats with a drop in spiritual malaise,activity and flexibility, hair gradually less yellow jersey, decrease into appetite,weight loss, thin pond or mucus blood stool.After14days drug treatment,HDG group rats general situation,mental state, activity and stool form werereturned to normal,with N of rats were similar.SASP group, MDG group andLDG group generally improved to varying degrees, but no HDG groupimproved obviously, the LDG group improvement worse than the MDG groupand SASP group.The DAI scores：HDG group（0.40±0.35）、MDG group（0.78±0.29）、LDG group（1.44±0.33）、SASP group（1.55±0.41）compared with M group（2.48±0.33）（P <0.05），the differences were statistically significant (P <0.05);The MDG group, LDG group, SASP group compared with HDGgroup，the differences were statistically significant (P <0.05);The LDG group,SASP group compared with the MDG group，the difference were statisticallysignificant (P <0.05); there was no statistically significant difference betweenSASP group and LDG group (P>0.05).2The general view and HE staining histopathological observation of colonicmucosaThe general view:HDG group was similar with N group,M group was theworst in all groups,SPAP group, MDG group and LDG group theimprovement of the situation of colonic mucosa were worse than HDG group,but better than M group,MDG group was a little better than SPAP group andLDG group.HE staining histopathological observation:Compared with N, M grouppresented numbers of inflammatory cells infiltration,crypt abscesses,glanddamage,goblet cells decreased and hyperemia edema, erosion and ulcerationof different level.HDG was similar with N group,SPAP group, MDG groupand LDG group of colonic mucosa recovery were between HDG group and Mgroup,the MDG group was better than that of LDG group and SASP group.3The content of TNF-α,IL-4and SOD in the serum of ratsTNF-α: Compared with N group,the serum TNF alpha level rised to different extent.The HDG group (0.63±0.09)compared with the Ngroup(0.55±0.11), there was no statistically significant difference(P>0.05);The treatment groups compared with M group (1.29±0.12), thedifference was statistically significant (P<0.05);MDG group (0.84±0.11),LDG group (0.89±0.29) and SASP group (1.14±0.12) compared with HDGgroup,the difference were statistically significant (P<0.05); LDG group andSASP group compared with MDG group, the difference were statisticallysignificant (P<0.05);LDG and SASP group, there was no statisticallysignificant difference (P>0.05).IL-4: Compared to N group (1.57±0.12), serum IL-4content of othergroups rats decreased, the difference had statistical significance;HDG group(1.41±0.15), MDG group (0.83±0.09), LDG (0.83±0.13mm) and SASPgroup (0.94±0.16) compared with the M group (0.54±0.09), the differencewere statistically significant (P<0.05);MDG group, LDG group and SASPgroup compared with HDG group，the difference were statistically significant(P<0.05);There was not statistically significant difference among SASP、MDG and LDG（P>0.05).By looking at the average, HDG group was betterthan that of the MDG group, LDG group and SASP group; There was noobvious difference among MDG group, LDG group and SASP group.SOD: Compared with N group (403.03±17.89),The content of SOD ofthe rest groups decreased, the difference were statistically significant (P<0.05);HDG group (388.60±15.67), MDG group (348.04±14.29), LDG (312.04±9.80mm), SASP group (341.34±12.88) content of SOD in serum of rats comparedwith M group (260.66±13.17) were higher, the difference had statisticalsignificance (P<0.05); MDG group, LDG group and SASP group compared tothe HDG group, the difference were statistically significant (P<0.05);Therewas not statistically significant difference between SASP group and MDGgroup（P>0.05)；there was statistically significant difference between SASPand LDG（P<0.05)；there was statistically significant difference between MDGand LDG（P<0.05),combining with the averages, the MDG was better thanthat of LDG. Conclusion:1The experiment proved that the LanYinFengYuHuaZhuoJieDuDecoction can not only improve the general situation of rats, promote thehealing of mucosa damage, and in a certain extent concentration-responserelationship, large dose group is better than middle and low dose group.2The mechanism of LanYinFengYuHuaZhuoJieDu Decoction in thetreatment of UC could be by lowering the serum TNF alpha proinflammatoryfactor, improve the level of IL-4and SOD anti-inflammatory factors,so as toachieve the effect of anti-inflammatory and immune regulation. |
