PurposeTo study the expression of YAP and β-catenin in human normal breast tissue,breast adenoma and breast infiltrating ductal carcinoma (not otherwise specified), analyze the relationship between the clinicopathological characters of breast infiltrating ductal carcinoma and two factors, further to explore their roles in breast infiltrating ductal carcinoma occurrence, development process and prognosis.MethodThe expression of YAP and β-catenin in 32 cases of human normal breast tissue,40 cases of breast adenoma and 97 cases of breast infiltrating ductal carcinoma was evaluated by immunohistochemical staining (streptavidine-peroxidase method).Result(1) The expression of YAP in human normal breast tissue, breast adenoma and breast infiltrating ductal carcinoma was decreased gradually, which in normal breast tissue was higher than that in breast adenoma and breast infiltrating ductal carcinoma. It provided a statistically significant result (P<0.05). Positive expression of β-catenin was remarkable higher in breast adenoma and breast infiltrating ductal carcinoma than that of normal breast tissue. in human normal breast tissue, breast adenoma and breast infiltrating ductal carcinoma, the positive expression rate was increase in turn, which provides the statistically significant difference(P< 0.05).(2)The expression of YAP was no statistically correlate with the age,histological grade, tumor size, lymph node metastasis, menopausal status of patients, yet associated with triple-negtive breast cancer (P<0.05), There were no correlation between the expression of β-catenin and breast infiltrating ductal carcinoma patients’ age, tumor size, menopausal status, etc(P>0.05),but the expression of β-catenin was statistically correlate with histological grade, lymph node metastasis and triple-negtive breast cancer(P<0.05).(3) The positive expression of YAP and infiltrating ductal carcinoma of the breast of overall survival in patients with no correlation (P>0.05),Which was associated with relapse-free survival rate (P< 0.05).The positive expression of β-catenin was no statistically correlate with overall survival rate and relapse-free survival rate (P>0.05).Yet YAP (-)/β-catenin (+) associated with overall survival rate, and relapse-free survival rate (P<0.05).(4)The expression of YAP and β-catenin were all negatively relative in breast adenoma and breast infiltrating ductal carcinoma (rs negative, P<0.01).These two proteins were not relative in human normal breast tissue(P>0.05).Conclusion:(1) The expression level of β-catenin in breast infiltrating ductal carcinoma was frequently upregulated,and the positive expression of β-catenin was statistically correlate with lymph node metastasis.It suggests that β-catenin plays an important role in acceleratting the occurrence and the metastasis of breast infiltrating ductal carcinoma.(2) The positive expression of β-catenin was statistically correlate with histological grade in breast infiltrating ductal carcinoma.It indicates that β-catenin can be used as a reference designator to estimate the tissue classification of breast infiltrating ductal carcinoma.(3) The expression of YAP and β-catenin was negative correlation in breast infiltrating ductal carcinoma (non specific).YAP (-)/β-catenin (+) associated with overall survival rate, and relapse-free survival rate (P<0.05).Combinded detection of YAP with β-catenin help the clinical diagnosis of breast infiltrating ductal carcinoma and prognosis.
