Buckwheat is a higher nutritional value of coarse grain crops, which contains a variety of flavonoids compounds has a role in the treatment of bronchitis, coronary vasodilation, hypoglycemic, anti-cancer. There are many literatures have reported the development of buckwheat,such as the extraction method of flavonoids in buckwheat, comparison of flavonoids content in buckwheat germination, the application of flavonoidsin Tartary Buckwheat, the improvement of nutritional beverage of Tartary Buckwheat, the nature and flavonoid antioxidant of buckwheat, the mechanism of all kinds of chemical reaction, method for determination of total flavonoids in Tartary buckwheat. However, four kinds of main flavonoids indifferent batches of rutin, kaempferol-3-rutinoside, quercetin and kaempferolhavebeen rarely reportedby using high performance liquid chromatography. And these research samples collected were not comprehensive, and have been limited to a single region, the comparison and anal-ysis of samples were single, and the describes of some methods for the determination. The difference was not involvedin the main producing areas of tartary buckwheat flavonoids composition and content. In this experiment, by using high performance liq-uid chromatography, the determination of 21 are different as such as Sichuan, different batches of four kinds of main flavonoids have been compared and analysed such asrutin, kaempferol-3-rutinosidein different varieties of buckwheat seeds powder and bran.Research methods:Using reversed-phase HPLC in 21 kinds of buckwheat, with chromatography column of Damonsil C18(4.6mm × 250 mm,5 μm),mobile phase of acetonitrile-0.2% acetic acid solution, flow rate of 1.0 mL/min, detection wavelength of 354nm, and column temperature of 30 ℃. Test results showed,21 kinds of buckwh-eat seeds of different varieties in room temperature detection of the moisture contentin 4.908%~6.994%. Branquality in thecondition of constant weight, mass fraction of 21.84%~30.03%, accounting for 1/4 of the quality tartary buckwheat bran weight. The linear range of rutin was 20~400g/mL. The linear range of the kaempferol-3-rutinoside was 2-40 g/mL, the linear range of quercetin was 1~20 g/mL. Different varieties of buckwheat seed powder, rutin content was 12.14~20.32 mg/g, the content of the kaempferol -3-rutinoside was 1.479~2.872mg/g, the content of quercetin was 0.0146~0.9014mg/g, kaempferol content was 0~0.0159mg/g. The four major flavonoids were the determination of the total content in the 14.02 ~22.87 mg/g; flavonoids contentin buckwheat bran content less than the overall seed powder, the content of rutin range was 4.269~12.12 mg/g, the content of kaempferol-3-rutinoside was 0.2585~1.015 mg/g, the content of quercetin was 0.0618~0.7354 mg/g, the content of kaempferol was 0~0.0263 mg/g, four kinds of content total flavonoids was 4.651~13.15 mg/g.Through the determination of research, quercetin-3-rutinoside (rutin) content was found the highest in the seed powder and bran, up to 90%, followed by kaempferol-3-rutinoside and quercetin, but the content of kaempferol in 1/10000 of buckwheatmass, was the lowest one, and some samples for content the detection limit was from kaem-pferol. With the determination latest collection offlavonoidcontent in Buckwheat of 21 varieties of seeds powder and buckwheat bran, and provides, the reliable dateswere gotten for the development and utilization, the buckwheat seed powder dates provide reliable evidence for the breeding of fine varieties of buckwheat seeds.Many thesis have reported Euphorbia lathyris L. medicinal value and the chemical composition and the contentoffatty oil, terpenoids and other chemical constituents. The irritating ingredients and the anticancer components of terpenoids have been elucidated. However, there are few reports of its anticancer mechanism, and comprehensive utilization is not enough, especially in the study of multi-drugresistance of cancer. Euphorbia factorL1、Euphorbia factorL3 of Euphorbia factor L1-L11 have been found the obvious anti-tumor activity, but its anticancer activity is still not strong enough. According to reports of thesis:Change the parent nucleus substituent influenceits spatial structure is likely to increase its activity, and achieve the purpose of drug development. The concern of this reports are rare. This study describes the anticancer activity of Euphorbia factor L3isolation, structure modification, synthesis of derivatives, havebeen determined the chemical structure of derivatives, new compounds with potential drug properties obtained.Conclusion:the four major flavonoids of 21 different batches, different producing areas of tartary buckwheathave been determinated and analysed by using high performance liquid in this thesis. Not only evaluated flavonoid content in tartary buckwheat, and discusses the quality, but also provides a scientific basis for the control of the quality standard of the development of tartary buckwheat; Euphorbia factor L3 extraction and structure transformation, affected the spatial structure, selected compounds with activity, has provided the scientific basis for the research of anticancer drugs. Synthesis method is fast, accurate, good repeatability by using natural products as lead compounds for new drug, provided a theoretical basis for the medicine, for future pharmacological testing activity screening, and determining the structure-activity relationship, Euphorbia factor L3 further structural modification.
