Objective: To explore the impact of Chinese medicine Xuan Fei Yang Yin Bu Shen decoction on the intestinal peristalsis function and the magnitude of average optical density of vasoactive intestinal peptide(VIP) and substance P(SP) in the myenteric nerve plexus, study the mechanism of releasing lungs kidney Yin decoction on chronic constipation consequently, by means of establish the model of constipation mice.Methods: 70 SD rats were randomly divided into 7 groups: blank group, model group, mosapride group, Xuan Fei Yang Yin Bu Shen decoction(high-dose group, middle-does group, low-dose group). The model group and control group were intragastric administration with compound diphenoxylate to establish constipation model. After the success of the copy of constipation model, the control groups were intragastric administration with diphenoxylate, Xuan Fei Yang Yin Bu Shen decoction(high-dose, middle-does, low-dose) and maren boluses respectively. The rats were executed after two weeks. Determine the alvine pushing rate of ink before execute the rats. Determine the magnitude of average optical density of VIP and SP by immunohistochemical staining.Results: The shit grain number in 24 hours and the wet weight of stools in 24 hours has significant difference(p<0.05) compare the blank group with the model group, the traditional chinese medicine group, mosapride group, maren boluses group.Immunohistochemistry results:(1) SP: Compared with the control group, the SP were significantly reduced in model control group, has a significant statistical significance(P <0.01); the magnitude of average optical density are increase distinctly in the traditional chinese medicine group, mosapride group and maren boluses group, had statistical significance. We can find that the traditional chinese medicine, mosapride, maren boluses can improve the the magnitude of average optical density of SP which in the myenteric nerve plexus remarkably; In addition, the increases of average optical density value of SP is more apparent in high dose group rats rather than mosapride group, Chinese traditional medicine(middle and low dose group) and the maren pills group(P < 0.05). The increase of the average optical density value of SP in colon in the middle-dose group of Traditional Chinese medicine(TCM) is higher than those of the low-dose group of traditional Chinese medicine(TCM) and the maren pills group(P < 0.05). Compare the middle-dose group of Traditional Chinese medicine(TCM) with the mosapride group, the increase of the average optical density value of SP in colon has no significant difference(P > 0.05). The increase of the average optical density value of SP in colon in the mosapride group is more obvious rather than the low-dose group of Traditional Chinese medicine and the maren pills group(P < 0.05). Compare the low-dose group of Traditional Chinese medicine with the maren pills group the increase of the average optical density value of SP in colon has no significant difference(P > 0.05). The increase of the average optical density value of SP in Traditional Chinese medicine group has dose-effect relationship with the concentration of the decoction.(2)VIP: The average optical density value of VIP: Compared with the blank group, the average optical density value of VIP in model group increased significantly, has significant statistical significance(P < 0.01); the magnitude of average optical density of VIP are decrease distinctly in the traditional chinese medicine group(high-dose, middle-does, low-dose), mosapride group and maren boluses group after three weeks treatment, and has statistical significance(P<0.05). The traditional Chinese medicine, mosapride, maren boluses can decrease the magnitude of average optical density of VIP which in the myenteric nerve plexus remarkably. The decrease of the average optical density value of VIP in Traditional Chinese medicine group has dose-effect relationship with the concentration of the decoction.Conclusion: The increase of VIP in the myenteric nerve plexus can loosen the intestinal smooth muscle and restrain intestinal peristalsis, and then generate the constipation. The decrease of SP in the myenteric nerve plexus may be lead to constipation by recede the secretion function of gastrointestinal tract. Xuan Fei Yang Yin Bu Shen decoction can decrease the the magnitude of VIP and increase the content of SP in the myenteric nerve plexus of colon, which may be one mechanism of it cure the constipation. It propose that the function of purgation of Xuan Fei Yang Yin Bu Shen decoction is relate to regulation of neurotransmitter.
