The Effect Of Xuan Fei Bu Shen Yang Yin Decoction On 5-HT₃R?eNOS And Neurofilament Protein In Rats With Slow Transit Constipation

Objective: To explore the possible mechanism of Xuan Fei Bu Shen Yang Yin decoction to treat STC by investigating the content of 5-HT3R?e NOS and neurofilament protein in the colonic tissue of rats with slow transit constipation,and to provide the reference data for the treatment of STC with Chinese medicine.Methods: Sixty SD rats were randomly assigned into two groups:the blank group with ten rats and the model group with fifty rats.Rats in the model group were given compound diphenoxylate aqueous solution through administration by gavage,while rats in the blank group were intragastric administration of normal saline.After successfully molding,rats in the model group were randomly divided into five groups.Rats in the model group and the blank group were given normal saline by intragastric administration,and the other groups were given the homologous drugs to treat them.Two weeks later,all the rats were putted to death after they were intragastric administration of ink.At last,the whole tharm of rats were disemboweled to measure the enteric driving rate,and the content of 5-HT3R?eNOS and neurofilament protein in the proximal colon of rats were detected with immunohistochemistry technology after the specimens were collected.Results:(1)The driving rate of carbon powder in the enteric canal of rats in the model group obviously reduced in comparison to the blank group on a statistically significant difference(P<0.01).The driving rate of carbon powder in the enteric canal of rats obviously improved in the Cisapride group and all the groups of traditional Chinese medicine in comparison to the model group on a statistically significant difference(P<0.05).(2)The content of 5-HT3 R in the colon of rats increased in the Cisapride group and all the groups of traditional Chinese medicine in comparison to the model group on a statistically significant difference(P<0.05).(3)The content of NF200 in the colon of rats decreased in the Cisapride group and the high and middle groups of traditional Chinese medicine in comparison to the model group on a statistically significant difference(P<0.05).(4)The content of eNOS in colon of rats barely changed in the Cisapride group and all the dose groups of traditional Chinese medicine in comparison to the model group on a statistically non-significant difference(P>0.05).Conclusion: The mechanism of Xuan Fei Bu Shen Yang Yin decoction treating STC may be by increasing the content of 5-HT3 R and decreasing the content of NFP in the colon,while promoting intestinal peristalsis,but there was hardly any effect on the content of eNOS in the colon.
