Objective:To study the correlation of P120catenin(P120ctn) and RhoA in normal cervix, precancerous lesions of uterine cervix tissue and cervical carcinoma tissue by detecting the expression level of P120ctn and RhoA. And to investigate the role of P120ctn and RhoA in the occurrence and development of cervical cancer.Methods:Immunohistochemistry (IHC) Envision was used to measure the expression of P120ctn and RhoA in 13 cases of normal cervical tissues,16 cases of precancerous lesions of uterine cervix tissues (including 8 cases of cervical intraepithelial neoplasia grade Ⅱ and 8 cases of cervical intraepithelial neoplasia grade Ⅲ),45 cases of squamous carcinoma and 15 cases of adenocarcinoma of cervix. The chi square test was used to analysis the expression of P120ctn and RhoA in different cervical tissues and their relationship with various clinical pathological parameters. And the correlation between the expression of P120ctn and RhoA in cervical cancer was tested by Spearman rank. The two-sided test was applied in all of these results, P<0.05 was statistically significant.Results:1. P120ctn was mainly located in the cell membrane of normal cervical tissues, precancerous lesions of uterine cervix tissues and Cervical cancer tissues, Some ectopic expression in the cytoplasm; 2. The abnormal expression rate of P120ctn in normal cervix, precancerous lesions of uterine cervix and cervical carcinoma were 53.84%(7/13), 81.25%(13/16),90.00%(54/60), respectively, and statistical significance was existed among the three groups (χ2=8.664, P=0.008). There was no statistical significance between normal cervix group and precancerous lesions of uterine cervix group(χ2=2.537, P=0.226). there was no statistical significance between precancerous lesions of uterine cervix group and cervical cancer group (χ2=0.840, P=0.387)). The statistical significance was found between normal cervix group and cervical cancer group (χ2=8.288, P=0.005); 3.In cervical cancer group, the expression of P120ctn was significantly collected with lymph node metastasis, depth of stromal invasion (P<0.05); However, no statistically significant difference was found in parametrial infiltration, vascular in filtration, tissue types, differentiation,patients’ FIGO stage, patient’s age and tumor size (P>0.05);4. It is found that RhoA expression was mainly located in the cytoplasm of normal cervical tissues, precancerous lesions of uterine cervix tissues and cervical cancer tissues;5. The positive expression rate of RhoA in normal cervix, precancerous lesions of uterine cervix tissues and cervical carcinoma were 0.00%(0/13)、50%(8/16) and 61.66%(37/60), respectively, and statistical significance was existed among the three groups (χ2=18.270, <0.001). However, There was statisticalsignificance between normal cervix group and precancerous lesions of uterine cervix group(χ2=11.981, P=0.003). there was no statistical significance between precancerous lesions of uterine cervix group and cervical cancer group (χ2=0.703, P=0.409)). The statistical significance was found between normal cervix group and cervical cancer group (χ2=21.305, P<0.001);6. In cervical cancer group, the expression of RhoA in patients was significantly collected with lymph node metastasis and differentiation degree and vascular infiltration (P<0.05); however, there was no statistical significance in the FIGO stage, parametrial infiltration, depth of stromal invasion, tissue types, tumor size and patient’s age (P>0.05);7. There was correlation between the expression of p120ctn and RhoA in cervical cancer (rs=0.341, P=0.008)Conclusion:1. The abnormal expressions of P120ctn and positive expressions of RhoA showed an increasing trend among normal cervical tissue, precancerous lesions of uterine cervix and cervical carcinoma. Both of them may be associated with the development of cervical cancer. 2. The abnormal expression rate of P120ctn in cervical cancer with lymph node metastasis and depth of myometrial invasion group were higher than the control group; but it was not associated with parametrial infiltration, vascular in filtration, tissue types, differentiation,patients’FIGO stage, patient’s age and tumor size.3.The positive expression of RhoA in cervical cancer with lymph node metastasis group,parametrial infiltration group and vascular invasion group were higher than the control group; but it was no associated with the FIGO stage, parametrial infiltration, depth of stromal invasion, tissue types, tumor size and patient’s age.4. There was correlation between the expression of P120ctn and RhoA in cervical cancer patients.
