ObjectiveTo explore the effects and mechanisms of the decrease of ARTN on proliferation and invasion and metastasis of gastric carcinoma cells. And to detect the expression of ARTN gene in gastric carcinoma. And to confirm the relationship between the ARTN gene and the occurrence, development of gastric cancer combine with the clinical and pathological characteristics of the patients.MethodsWestern Blot were used to detect the expression of ARTN gene in gastric cancer cell line of SGC7901,MGC803, BGC823, and select the highest expression one. Three kinds of siRNA(1808, 1383 and 1604) were synthesised to interference the gene of ARTN, Gene transfection were used to transfer the si RNAs into the gastric cancer cell line. Then flow cytometry was used to observe the transfection efficiency. Real-time PCR and Western Blot were used method to detect the expression of protein and mRNA of ARTN after the interference, and select the group that has the highest efficiency of silence the ARTN. CCK-8 was used to detect the cell proliferation, and the transwell chamber was used to detect cell migration and invasion. Real-time quantitative PCR and Western Blot were used to detect the difference of expression of gastric cancer related gene(P53, MMP-2, MMP-9, VEGF and BCL-2) in the level of RNA and protein before and after interference. Immunohistochemistry and Western Blot were used to detect the expression of both gastric cancer tissue and normal gastric mucosa.ResultsARTN gene was most highly expressd in MGC803.Group 1383 had the highest silencing efficiency on interfering the expression of ARTN. The expression of ARTN in RNA and protein levels were significantly decreased afer transferred with siRNA. After the interference of ARTN gene, the ability of cell proliferation, migration ability and invasion of MGC803 were significantly decreased, and the expression of P53, MMP-2, MMP-9, VEGF and BCL-2 were reduced in RNA and protein levels. The expression of ARTN in gastric carcinoma were higher than in the gastric mucosa.ConclusionARTN gene could promote the proliferation, migration and invasion of gastric cancer cells, and reduce the expression of gastric cancer-related gene. It is related with differentiation of gastric cancer, depth of invasion, metastasis and clinical stage, and can be used to evaluate the clinical stage and guide the treatment. It plays an important role in the development and pathogenesis of the gastric cancer.
