The content of this paper derived from the national natural science fund projects " The model effects of Radix Scutellariae and Gardenia on processing compatibility in Huangqin qingfei decoction". Huangqin qingfei decoction, come from Volume 17 of Yuan’s The Wei Sheng Bao Jian created by Luo Tianyi, which is mainly composed of Radix Scutellariae and Gardenia. The prescription has a big difference in various regions. According to the clinical efficacy of Huangqin qingfei decoction, the compatibility of radix scutellariae and gardenia, including raw Radix scutellariae and wine Radix scutellariae, gardenia and Stir-baked Fructus Gardenia, was carried out by HPLC fingerprint, LC-MS/MS, IR and multifaceted quantitative research which studied chemical changes about terpenoids, flavonoids, diterpenoid pigments, phenolic acids systematically. The study provided scientific basis for clinical reasonable application of Huangqin qingfei decoction. Details are shown as follows:·Chapter 1 Literature researchLiteratures about radix scutellariae and gardeniae were reviewed around the research content. We has analyzed and summarized the literature on chemical composition the clinical application of Huangqin qingfei decoction, pharmacological effects of gardenia and radix scutellariae.Chapter 2 Preparation of samples used in the compatibility of Huangqin qingfei decoctionRaw herbal materials of gardenia and radix scutellaria in accordance with China pharmacopoeia were collected contains as raw material medicine, accordance with the Preparation Method, pilot batch preparation of raw gardenia and focal gardenia, raw radix scutellaria, wine radix scutellariae and by the orthogonal experiment design, prepare the samples of the compatibility different decoction pieces.Chapter 3 Separation and Purification of iridoid glycosides from Gardenia jasminoides FruitSeparation and Purification of six iridoid glycosides from the Extract of Gardenia jasminoides Fruit Using Medium-pressure Liquid Chromatography (MPLC). The analysis of these compounds was confinned by high-performance liquid chromatography (HPLC) and chemical structures were elucidated by 1H-NMR,13C-NMR. Macroporous resin adsorption combined with the preparation (MPLC) separation and HPLC chromatographic technologies were used for rapid purification iridoids from 30% ethanol elution, which provided reference substance for the quantitative research. Chapter 4 Study on HPLC fingerprint of Huangqin qingfei decoction and identification of main chromatographic peakMethods of HPLC fingerprint with two wavelengths (254nm,440nm) were established in multiple fingerprint analysis for Huangqin qingfei decoction. The results showed that HPLC overall figure was not significant difference in four kinds of compatibility mode under 254nm, but has obviously differences under 440nm.20 chromatographic peaks were identified under 254nm with reference substance while four diterpenoid ingredients were identified at 440nm. This study reveals chemical composition and its change rule of the compatibility between radix scutellaria and gardenia preliminarily.Chapter 5 Ingredients identification in compatibility of radix scutellaria and gardenia by LC-MSMain compositions were identified in the compatibility of radix scutellaria and gardenia according to the positive and negative ions by HPLC/ESI-MS detection method. Results showed that 32 compounds including 9 of iridoid glycosides, 6 of diterpenes, 1 of organic acids, 16 of flavones glycosides were identified. It is provided an important reference for further research on chemical composition and new compounds of radix scutellaria and gardenia and established the scientific basis for change law of the chemical compositions in Huangqin qingfei decoction.Chapter 6 The main ingredients quantitative study on the compatibility of radix scutellariae and gardeniaThe content of 24 kinds of ingredients in the compatibility of radix scutellariae and gardenia was determined by HPLC under two detection wavelength at the same time. The content of iridoid glycosides and flavonoids ingredients were higher in the combination compatibility of radix scutellariae and gardenia than exclusive use under 254nm. The result of iridoid glycosides compounds were obviously. The content, proportion and types of diterpenoid compounds which come from gardenia mostly were different varieties under 440nm. Diterpenoids in Gardenia have a enormous change after changing into Stir-baked Fructus Gardenia, The content of diterpenoid were higher in the combination compatibility of radix scutellariae and gardenia than used separately under 440nm. This study illustrated the varieties connotation on the compatibility of radix scutellaria and gardenia.Chapter 7 The study on compatibility of radix scutellaria and gardenia by infrared spectrumPrincipal component changes were speculated by comparing radix scutellariae, fructus gardeniae and the compatibility of radix scutellariae and gardenia of infrared spectrum including forst order spectrum, second derivative spectra and second-order spectrum. According to the different characteristics absorption of every peak by the stretching vibration frequency, we can infer the types of chemical structures. Furthermore, the change rule of compatibility composition can provide a validation for the identification of composition by LC-MS/MS. Infrared spectrums of prescription were not the simple sum of the infrared spectrum radix scutellaria and gardenia. We presumed chemical environment of compatibility had changed greatly.Chapter 8 Results and discussionThis paper interpretated chemical compositions and its change rule of compatibility (radix scutellariae, gardenia= 2:1) based on the literature research, HPLC fingerprint analysis and LC-MS. Qualitative identification of different compatibility study on compatible proportion were through infrared chromatographic. This study expounds the chemical components of the compatibility between radix scutellariae and gardenia and its change law and illustrates the many ingredients, multicomponent of compatibility between radix scutellariae and gardeniae. It illustrates basic connotation of the compatibility of radix scutellariae gardenia and provides the beneficial reference and reliable clues for quality evaluation standard and its clinical rational use.
