Objective: To establish the HPLC fingerprint of Artemisia rupestris.L different extracts,according to the result of HPLC fingerprint and its antiviral activity,"spectrum-effect" related,we can analysis the chemical composition of the antiviral activityto reveal the material base of antiviral effect of Artemisia rupestris.L.Methods:① To study extraction method of Artemisia rupestris.L Flavonoids and HPLC chromatographic conditions,traditional medicine fingerprint technology is used to establish the HPLC fingerprint of Artemisia rupestris.L,similarity is used as criteria to standardize characteristic peak and determine the similarity evaluation results;②We evaluated anti-influenza virus activity of Artemisia rupestris.L by MDCK cell model and the Cytopathic effect (CPE),combine with MTT method;We evaluated anti-hepatitis B virus activity by cell model of HepG2.215;③By using the gray modeling software,we combined HPLC fingerprint data with antiviral results to get correlation coefficient between chromatographic fingerprint features of Artemisia rupestris.L different extracts and their antiviral effect,determined the order of contribution of composition for"anti-influenza,anti-hepatitis B virus";④Finally,using the method of HPLC-MS,we understanded which chemicals might be the substances of Artemisia rupestris.L’s antiviral activity.Results:①Building HPLC fingerprint of 20 extracts of Artemisia rupestris.L, through the "Chromatographic fingerprint similarity evaluation system",we determined nine common chromatographic peaks;②Using vitro antiviral test,in different extraction parts samples,S2,S5 and S6 showed anti influenza virus activity;S2,S3,S4,S5 S7,S8,S11,S12, S13, S14,S16 showed anti-hepatitis B virus activity; ③Through the gray correlation analysis,we determined the order of various chemical composition contribution to the anti -influenza virus activity:9>4>3>l>7>2>6>5>8;the order of various chemical composition contribution to the anti-hepatitis B virus activity: 9>7>5>3>4>2>8>1>6.④ Through the HPLC-MS method,the main active ingredient of anti-influenza virus of Artemisia rupestris.L may be casticin、rupestonic acid、apigenin.etc; the main active ingredient of anti-hepatitis B virus of Artemisia rupestris.L may be casticin、acacetin.etc.Conclusion:The HPLC fingerprint method of Artemisia rupestris.L different extracts is simple,reproducible;the method using the model of "spectrum-effect" of Artemisia rupestris.L extracts antiviral to evaluate antiviral activity is accurate and feasible,its results are more scientific、comprehensive,actual.
