ObjectiveTo Study the expression of placenta growth factor and matrix metalloproteinase-2inthe placenta of patients with preeclampsia,in hopes of making the mechanism of PE moreclear.MethodsAs the experimental groups,40placenta specimens of PE were collected from theobstetrical department of Affiliated Hospital of Tai Shan Medical College as theexperimental group. The control specimens were placenta tissue of normal-term cesareandelivery.The diagnosis standard of PE patients refers to the seventh edition of obstetricsand gynecology.The expression of PLGF and MMP-2was detected by immunohist-ochemistry methods.Results1. The cellular localization of PLGF and MMP-2in placenta from normal villilobules was mainly located in the syncytiotrophoblast and cytotrophoblast. Positivestaining areas were located in cytoplasm.2. The cellular localization of PLGF and MMP-2in preeclampsia placenta weresimilar to that of the normal placenta tissues.The positive staining areas were located incytoplasm.Positive staining distribution was consistent with normal placenta, but the colorwas lighter than the control group.3. The positive rates of MMP-2expression in the normal group,the mildpreeclampsia group and the severe preeclampsia group were:93.3%,70.6%,39.1%,respectively.Comparing the mild preeclampsia group and the normal group, the resultsshow no significant differences(P>0.05).Comparing the severe preeclampsia group and thenormal group, the results indicate significant differences(P<0.05).Comparing the mildpreeclampsia group and the severe group, the results indicate significant differences(P<0.05).4.The positive rates of PLGF expression in the normal group,the mild preeclampsiagroup and the severe preeclampsia group were:53.3%,41.2%,13.0%,respectively.Compared with the mild preeclampsia group results, the normal results show no significantdifferences(P>0.05).Compared with the severe preeclampsia group results, the normalgroup results were significantly different(P<0.05).Comparing the mild preeclampsia groupand the severe group, the results demonstrated significant differences(P<0.05).5.The expression of PLGF in the severe preeclampsia group showed positivecorrelation with the expression of MMP-2(r=0.697,P<0.05).ConclusionThe expression of MMP-2and PLGF in preeclampsia placenta appeared up-regulatedin PE. This demonstrates that these two factors may be involved in the pathogenesis of PEby means of affecting shallow placenta implantation in preeclampsia,ischemia,hypoxia andby means of apoptosis of trophoblasts,injury of vascular endothelial cells and arteryvasospasm.This may prove to be useful in understanding the prediction and treatment ofpreeclampsia.
