Objective:To the detection of uterine leiomyoma tissues and tumor adjacentnormal tissues of uterine muscle of estrogen receptor (ER), progesteronereceptor (PR), matrix metalloproteinase1(MMP-1) and apolipoprotein1(apoptosis related gene, Fas) gene polymorphisms, to explore thecorrelation between the onset of uterine leiomyoma in ER, PR, MMP-1and Fas gene polymorphism and the Northeast in the area of moleculargenetics.Methods:Collected in department of Obstetrics and Gynecology, SecondAffiliated Hospital of Jilin University, from2013June to2014August,200cases of hospitalized patients operation resection of uterine fibroids,divided into the uterine fibroids group (experimental group)200casesand tumor adjacent normal uterine muscle group (control group)200cases. Two groups of specimens are in operation excision immediately putinto liquid nitrogen frozen, to be measured. Using polymerase chainreaction restriction fragment length polymorphism (PCR-RFLP)technique, the susceptible gene of ER detection (rs2234693, RS9340799),PR (rs1145460, RS11224567), MMP-1(rs494379) and Fas (rs2234767,RS1800682) single nucleotide polymorphism (SNPs).Results:1.Comparison of rs9340799, rs2234693gene polymorphism, eachgroup of estrogen receptor (ER) Compared with the control group, the experimental group ofestrogen receptor (ER) rs9340799(A/G) no mutation, no significantdifference (P>0.05); rs2234693(C>T) mutation, the difference wasstatistically significant (P<0.05).2. Comparison of rs11224567, rs1145460gene polymorphism, eachgroup of progesterone receptor (PR)Compared with the control group, the experimental group ofprogesterone receptor (PR) rs11224567(C/T)no mutation, no significantdifference (P>0.05); rs1145460(C>/T)mutation, the difference wasstatistically significant (P<0.05).3. Comparison of rs494379gene polymorphism, each group of matrixmetalloproteinase1(MMP-1)Compared with the control group, the experimental group of matrixmetalloproteinase1(MMP-1) rs494379(A>G)mutation, the differencewas statistically significant (P<0.05).4. Comparison of rs2234767, rs1800682gene polymorphism, each groupof apoptosis related gene FasCompared with the control group, the experimental group ofapoptosis related gene Fas rs2234767(A>G), rs1800682(C>T) aremutated, the difference was statistically significant (P<0.05).Conclusion:1. The experimental group of estrogen receptor (ER) rs9340799(A/G) and progesterone receptor (PR) rs11224567(C/T) no mutation,illustrate the onset of uterine leiomyoma crowd gene polymorphism maybe associated with the northeast area of the two independent.2. The experimental group of estrogen receptor (ER) rs2234693(C>T), progesterone receptor (PR) rs1145460(C>/T), matrixmetalloproteinase1(MMP-1) rs494379(A>G), apolipoprotein rs2234767 (Fas)1(A>G), apolipoprotein rs1800682(Fas)1(C>T)5loci mutations,that incidence of uterine leiomyoma associated gene polymorphism maybe associated with the northeast area the5loci, play an important role inthe pathogenesis of uterine leiomyomas.
