Objective: To investigate the relationship of cell apoptosis andnonneoplastic epithelial disorders of vulva (NNEDV) byimmunohistochemical method, which is used to detect the expressionaldifference and correlation in apoptosis-related proteins Bcl-w and Beclin1inNNEDV, and provide scientific basis for the pathogenesis and treatment ofNNEDV study. Methods:We collectted samples with vulvar lesions (lesiongroup) and vulvar skin adjacent to the lesions (adjacentcy group) from42NNEDV patients and embed it in paraffin.We detected the number of apoptoticcells in lesion group and adjacency group using Tunel Apoptosis Assay Kit, inorder to make sure the different apoptosis index between the two groups. Thepositive expression of apoptosis-related proteins Beclin1and Bcl-w weredetected in the two groups by peroxidase-labeled rabbit two stepimmunohistochemical staining kit. The statistical analysis for mean opticaldensity of positive cells and the expressional difference and correlation ofBeclin1and Bcl-w in the two groups are made by ImagePro6.0which is aprofessional image analysis system. Statistical methods: experimental data wasprocessed by SPSS20.0statistical software. It had statistical significancewhen P<0.05. Results:(1) VSH lesion group and adjacency group weredifferent in apoptosis percentage (P<0.05), it was statistically significance. VLS lesion group and adjacency group were different in apoptosis percentage(P<0.05), it was statistically significance. It wasn’t statistically significantwhen mixed type lesion group and adjacency group are different in apoptosispercentage (P>0.05).(2) The expressional level of Beclin1was statisticallydifferent between VSH lesion group and adjacency group (P<0.05), and theexpressional level in lesion group was lower than in adjacency group. Theexpressional level of Beclin1was not statistically different between VLSlesion group and adjacency group (P>0.05), as well as between mixed typelesion group and adjacency group (P>0.05). The expression of Beclin1was notsignificantly deferent among VSH, VLS and mixed type lesion group (P>0.05).(3) The expression level of Bcl-w was statistically deferent between VSHlesion group and adjacency group (P<0.05), and the expression level in lesiongroup was higher than in adjacency group. The expression level of Beclin1was not statistically deferent between VLS lesion group and adjacency group(P>0.05), as well as between mixed type lesion group and adjacency group(P>0.05). The expression level of Bcl-w in VSH lesion was significantlyhigher than in VLS and mixed type lesion group (P<0.05).(4) The expressionof Beclin1and Bcl-w showed no correlation in lesion group (r=0.017,P=0.944>0.05), as well as in lesion-adjacent group(r=0.101, P=0.671>0.05) inVSH. The expression of Beclin1and Bcl-w showed no correlation inlesion-adjacent(r=0.472, P=0.076>0.05) group, while positive correlation wasobserved in lesion group(r=0.527, P=0.043<0.05) of VLS. The expression of Beclin1and Bcl-w showed no correlation in lesion group (r=-0.070,P=0.882>0.05), as well as in lesion-adjacent group(r=-0.084, P=0.859>0.05) inmixed type lesion group.Conclusion:(1) The different apoptosis indexbetween lesion group adjacency group demonstrates that cell apoptosis was animportant factor affecting Nonneoplastic Epithelial Disorders occurs on vulvaepithelium.(2)The expression level of Bcl-w was statistically differentbetween lesion group and adjacency group (P<0.05), and the expression levelin lesion group was higher than in adjacency group. It demonstrated the effectof anti-apoptosis of Bcl-w had an protective effect on the survival ofpathological cells, which was favorable for the development of lesions. Theexpression of Bcl-w was not significantly different between VLS and mixedtype lesion group (P>0.05).(3)The expressional level of Beclin1wasstatistically different between lesion groups and adjacency groups (P<0.05),and the expressional level in lesion group was lower than in adjacency group.It can be speculated that Beclin1leads to autophagic cell death and plays a rolein blocking lesions.(4) In lesion groups of NNEDV,the positive correlationwas only observed in lesion group(r=0.527, P=0.043<0.05) of VLS,whichdemonstrated the cooperative relationship of Bcl-w and Beclin1in the processof occurring and developing in VLS.
