| Objective:Using randomized, case-control study, to observe the effect of Cake-separated Moxibustion on the Expression of the nf-kappa B> TLR2and TLR4content in TLR2/4-nf-kappa B signaling pathway of Atheroscl-erotic vulnerable plaque Rabbits.From the cellular and molecular level to explore the action mechanism of cake-separated moxibustion medicine stabling atherosclerotic vulnerable plaque. It will provide important experimental evidence for clinical rational use of cake-separated moxi-bustion intervention development of this disease.Methods:60New Zealand white rabbits,Male and female half,were divided into5groups:Blank group(control group),model group(AS model group),direct moxibustion group (AS model plus direct moxi-bustion),cake-separated moxibustion group(AS model plus cake-separateed moxibustion) and medicine group(AS Model plus atorvasta-tin),12rabbits per group.AS vulnerable plaque rabbit models were established by feeding high-fat diet plus arterial intimal injury of the ventral aorta with balloon inrabbits,vulnerable plaque Rabbits were treated with different way of direct and cake-separated moxibustion〠atorvastatin on the same acupuncture points.For eight weeks.Testing index and method:1.took carotid artery blood, determined TCã€TG’s content after treatment by enzymic method,determined HDL-Cã€LDL-C’s content by colorimetric method, determined apoA and apoB by electro-phoresis method;2.Changes of ultra-structures of ASvulnerable plaque rabbit were observed with electron microscope;3. Hematoxylin-eosin Staining detection AS lining and film thickness in vulnerable plaques;4.Using immunohistochemical technique to detect abdominal aorta TLR2and TLR4, the nf-kappa B grey value, observe its antigen immune activity;5.Then the mRNA expression of TLR2ã€NF-κ Bã€TLR4was detected in the atherosclerotic plaques of rabbits with in fluorescence quantitative PCR technology.Results:1.Blood fat content:Compared with model group, The serum HDL-C〠Apo-A content of each treatment group increased (P<0.05或P<0.01), TCã€TGã€LDL-Cã€Apo-B content decreased (P<0.05或P<0.01).2.AS vulnerable plaque ultra-structures:(1) the model group there were serious pathological changes in endothelium and subendothelium which were filled with foam-like cell containing a great number of lipid droplets and lipide,lesion center of cell necrosis, disintegration, lipid precipitation; In the internal elastic membrane and membrane of elastic fiber fracture collapse occurs, the insect damage change;To a large number of smooth muscle cells by membrane lining the migration and proliferation, cell arrangement disorder, and there is a lot of foam cells and fibrous connective tissue formation.Show that the model successfully.(2) Compared with the model group, it suggested that the aortic endothelial cells in the cake-separated moxibustion group and atorvastatin treated group were more intact; most of them were normal in forms; internal elastic membrane was continuous; their thickness was even or uneven; cells of smooth muscles in the medial membrane were relatively normal; mitochondrion edema,vacuous change and small quantities of lipid droplets could be seen occasionally.It show that three kinds of treatment can effectively protect the AS vulnerable plaques rabbit aortic endothelial cells.3.AS lining and film thickness in the vulnerable plaques:(1)compared with the blank group, model group in rabbit aortic intima thickness, thickness of the film thickness signifycantly higher (P<0.01).(2)compared with model group, cake-separated moxibustion group, the atorvastatin treated group and direct moxibustion group directly rabbit aortic intima in endometrium film thickness were significantly lower(P<0.01).(3)compared with direct moxibustion group,cake-separated moxibustion group and atorvastatin treated group in rabbit aortic intima thickness of the film thickness was significantly lower (P<0.01).(4)cake-separated moxibustion group and atorvastatin treated group, two groups of the membrane, membrane thickness has no obvious difference (P>0.05).4.Abdominal aorta TLR2and TLR4,the nf-kappa B grey value and its antigen immune activity:direct moxibustion group and atorvastatin treated group, cake-separated moxibustion group were made vulnerable plaques of atherosclerosis rabbit abdominal aorta TLR2and TLR4, the nf-kappa B grey value expression significantly lower (P<0.01orP<0.05), and cake-separated moxibustion group, the atorvastatin treated group is better than that of direct moxibustion group.5.AS the vulnerable plaques rabbit TLR2and TLR4mRNA expression, the nF-kappa B:direct moxibustion and atorvastatin treated group, cake-separated moxibustion group were made AS vulnerable plaques rabbit abdominal aorta TLR2and TLR4, the nF-kappaB mRNA expression significantly lower (P<0.01or P<0.05), and cake-separated moxibustion medicine group, the atorvastatin treated group is better than that of direct moxibustion group.Conclusions:1.Cake-separated Moxibustion has good accommodation to blood fat and lipoprotein metabolism.2.Cake-separated Moxibustion can reduce the AS rabbit endometrium vulnerable plaques, the film thickness, inhibit intimal hyperplasia, delay the further development of the AS.3.Cake-separated Moxibustion can change AS vulnerable plaques rabbit aortic endothelial cell damage, protect the endodermis, delay the further development of the AS.4.Cake-separated Moxibustion can inhibit rabbit TLR2ã€TLR4ã€nF-kappa B activity in atherosclerotic plaques, and mRNAexpression, have the effect of atherosclerotic plaque stability. |
