Objective: Observation of herbal cake separated moxibustion on Atherosclerotic Vulnerable Plaque in rabbits (vulnerable atherosclerotic plaque, VAP) effects of abdominal aortic PI3K and the expression of AKT, to explore the mechanism of moxibustion with medicinal cake inhibition of PI3K/AKT pathway and thus the stability of VAP.Methods:60New Zealand rabbits, randomly divided into blank group, model group, direct moxibustion group, moxibustion group and drug group, each group of12only. Given high fat feeding and balloon injury of abdominal aortic method, with the wild type P53gene, Chinese Russell’s viper venom and histamine drugs trigger, preparation of rabbit VAP model, at the same time for different intervention measures. After twelfth weeks, TG TC from artery blood test, LDL-C, HDL-C, Apo-A, Apo-B content, HDL-C/LDL-C value calculation; pathological observation and plaque morphology microscope HE staining; acoustic densitometry measurement of abdominal aorta intima, media thickness (IMT); TEM observation of rabbit abdominal aorta endothelial cell ultrastructure; immunohistochemistry detection of PI3K, AKT and TNF-alpha gray value, observation of activated antigen immune three expression; real-time fluorescence quantitative PCR detection of PI3KmRNA, AKTmRNA and TNF-alpha mRNA.Results:1. Blood lipid content:compared with model group, the treatment group serum TC, TG, LDL C, Apo B content were significantly lower (P<0.05or P<0.01), HDL-C, Apo-A content were significantly higher (P<0.05or P<0.01).2. AS ultrastructure of vulnerable plaques:(1) the model set of rabbit aortic endothelial and endothelial lesion severity, lower sample contains a large amount of lipid and foam cells, focal center of cell necrosis, disintegration, lipid precipitation; In the internal elastic membrane and membrane of elastic fiber fracture collapse occurs, the insect damage change; To a large number of smooth muscle cells by membrane lining the migration and proliferation, cell arrangement disorder, and there are a large number of foam cells and fibrous connective tissue formation, indicates that the model successfully;(2) compared with model group, every medicine cake moxibustion group, the drug moxibustion group and direct moxibustion group rabbit aortic endothelial structure is similar to that of normal form mostly, the continuous elastic membrane, thickness is uniform or not, basic normal smooth muscle cells, mitochondria edema, cavitation duality can be change and a small amount of lipid droplets, showed that three kinds of treatment can effectively protect the AS vulnerable plaques rabbit aortic endothelial cells.3. AS lining and film thickness in the vulnerable plaques:(1) compared with the blank group, model group rabbit aortic intima thickness, the lining of the film thickness increased significantly (P<0.01), the model of success;(2) compared with model group, every medicine cake moxibustion group, the drug group and moxibustion group directly rabbit aortic intima thickness, thickness of lining in the film were significantly lower (P <0.01), the difference is statistically significant, indicating that moxibustion and drugs can prevent the development of intimal hyperplasia;(3) compared with direct moxibustion group, every medicine moxibustion group and drug group rabbit aortic intima thickness decreased significantly (P<0.01), lining to reduce the thickness of membrane (P <0.05), the difference is statistically significant.4. Antigen activity showed:every medicine cake moxibustion group and drug group could increase AS the vulnerable plaques rabbit the expression of inflammatory factor grey value, and the effect is better than that of direct moxibustion group. 5. RT-qPCR display:direct PI3KmRNA expression of moxibustion group and drug group significantly decreased (P<0.05), and bread every drug moxibustion PI3KmRNA expression was not significantly decreased (P>0.05); Direct moxibustion group, every medicine cake moxibustion AKTmRNA, TNF alpha mRNA expression decreased obviously (P<0.01); Drug group AKTmRNA, TNF alpha mRNA expression decreased, but not obvious (P>0.05).Conclusion:1. Herbal cake separated moxibustion can effectively reduce the TC, TG content of VAP in blood, and can reduce the LDL and Apo-B in blood increases, the content of HDL increased, and Apo-A content, suggesting that the metabolism of herbal cake separated moxibustion on lipid and lipoprotein has good regulating effect2. Herbal cake separated moxibustion has protective effect on VAP in rabbit abdominal aorta endothelial cells.3. Herbal cake separated moxibustion could inhibit AKT activity inhibition of abdominal aorta, abdominal aortic AKTmRNA expression, suggesting that the cake separated moxibustion is through inhibiting the expression of AKT, and thus the stability of VAP...
