Objective: Studying the effect of cake separated moxibustion on the expression of RhoA/ROCK1,and LXR mRNA of vulnerable atherosclerotic plaque rabbits,and exploring the mechanism of cake-separated moxibustion in stabilizing vulnerable atherosclerotic plaque.Methods:70 rabbits were randomly divided into blank group(n=10)and surgical group(n = 60).The blank group was fed with normal diet,and the surgical group was fed with high fat diet,then perform abdominal aortic balloon injury after two weeks.There are 55 surviving rabbits were randomly divided into the model group,direct moxibustion group,cake-separated moxibustion group,inhibitor group and drug group,with 11 rabbits in each group.The methods of treatment were given direct moxibustion,cake separated moxibustion,ear vein injection of fasudil and oral atorvastatin calcium for a total of 8 weeks.After treatment,all groups use Ad5-p53 recombinant vector for gene transfection except the blank group,and Chinese spots and viper venom were given 48 hours and 24 hours before execution.ELISA was used to detect the levels of TC,TG,LDL-C,HDL-C,apo-A,apo-B,and CYP7A1 in serum;Real-time fluorescence quantitative PCR was used to detect the expression of RhoA mRNA,ROCK1 mRNA and LXR mRNA in the abdominal aorta.Results:1.Blood lipid: ELISA results showed that compared with the blank group,the content of TG,TC,LDL,apo-B and apo-B/apo-A in the model group was significantly increased,the levels of apo-A,HDL and HDL/LDL were significantly decreased(P<0.01).Compared with the model group,the content of TG,TC,LDL,apo-B and apo-B/apo-A in the treatment groups were significantly decreased(P<0.01,or P<0.05),but in the aspect of regulating the content of apo-B,cake-separated moxibustion has the best effect,the levels of apo-A,HDL and HDL/LDL were significantly increased(P<0.01,or P<0.05).Compared with the western medicine group,the content of apo-B in the cake-separated moxibustion group was significantly decreased(P<0.05).but there was no significant difference in other indexes(P > 0.05).The above results show that cake-separated moxibustion, direct moxibustion and Western medicine can effectively regulate blood lipid levels,and the effect of herbal cake-separated moxibustion is the best in regulating the content of apo-B.2.Expression of AS vulnerable plaque rabbits on RhoA,ROCK1 and LXR mRNA: Compared with the blank group,the expression of RhoA mRNA and ROCK1 mRNA in the model group was significantly increased(? < 0.01),and LXR mRNA expression was significantly decreased(? < 0.05);Compared with the model group,the expression of RhoA mRNA and ROCK1 mRNA was significantly decreased(? <0.05,? < 0.01),the expression of LXR mRNA in the cake-separated moxibustion group and Western medicine group was significantly decreased(? < 0.05,? <0.01),there was no significant difference between the direct moxibustion group and the inhibitor group(? > 0.05),but there was an upward trend.The above results show that cake-separated moxibustion can inhibit the expression of RhoA and ROCK1 mRNA,increase the expression of LXRmRNA and enhance the reversal of cholesterol,so as to stabilize the vulnerable plaques of AS.3.The content of serum CYP7A1 in AS vulnerable plaque rabbits: ELISA results showed that compared with the blank group,the content of CYP7A1 in the model group was significantly decreased(? < 0.05).Compared with the model group,the content of CYP7A1 in each treatment group was significantly increased(? < 0.05,?< 0.01);Compared with the direct moxibustion group,the content of CYP7A1 in the cake-separated moxibustion group and the western medicine group decreased(? < 0.05,? < 0.01),while the CYP7A1 content in the inhibitor group increased(? < 0.01).Compared with the western medicine group,the CYP7A1 content in the cake-separated moxibustion group had no significant difference.The above results show that moxibustion separated by medicinal cake can increase the expression of CYP7A1,and there is no significant difference between moxibustion and western medicine group,but the effect of direct moxibustion is better than that of moxibustion separated by medicinal cake.Conclusion:1.The cake separated moxibustion can regulate the blood lipid level of AS vulnerable plaque model rabbits;2.The cake separated moxibustion can inhibit the expression of RhoA /ROCK mRNA,promote the expression of LXR mRNA,increase the content of CYP7A1,enhance the reversal of cholesterol,stabilized AS vulnerable plaque.
