Objective:To observe the effect to HLP and AS rabbits treated by herbal-cake-separated -moxibustion in cell and molecular level and to analyze whether the anti-hyperlipemia rationale of the therapy is to protect the vascular endothelial cells.Method:75 rabbits were divided into 5 groups randomly:blank group,model group of HLP and AS,direct-moxibustion group,herbal-cake-separated -moxibustion group,herb group,15 rabbits each group。model rabbits of HLP and AS were built by immunity-injury-method,cholesterol of blood plasma (TC),triglyceride(TG),high density protein(HDL-C),low density protein (LDL-C),apolipoprotein A(apo-A),apolipoprotein B(apo-B),endothelin (ET-1),prostacyclin(6-keto-PGF1α),thremboxane B2(TXB2) are determined after treated;the nitric oxide(NO) is determined with the method of nitrate reductase;the expression of VCAM-lmRNA and E-sel mRNA are determined with the method of RT-PCR;the change of ultrastructure of endothelial cells of the aorta of the rabbits is observed with electron microscope。 Results:①Content of TC,TG,LDL-C,apo-B in herbal-cake-separated -moxibustion group,direct-moxibustion group and herb group is obviously decreased(P<0.01 ),and content of HDL-C,apo-A in those groups is obviously increased(P<0.05 or P<0.01 ).In regulating effect on TC of HLP and AS rabbits,each of herbal-cake-separated moxibustion group and herb group is better than direct moxibustion group(P<0.01 ),②Content of ET-1,TXB2,ratio of ET-1/NO,TXB22/6-keto-PGF1α in herbal-cake- separated- moxibustion group,direct-moxibustion group and herb group is obviously decreased(P<0.05 or P<0.01 ),and content of 6-keto-PGF1α is obviously increased(P<0.05 )。③There is a tendency that content of NO increases in order of model group,blank group,herbal-cake-separated-moxibustion group,directmoxibustion group,herb group,but there is no statistics difference between each two groups。④The expression of VCAM-1mRNA and E-selmRNA in herbal-cake-separated- moxibustion group,direct-moxibustion group and herb group is obviously decreased(P<0.05 or P<0.01 )。⑤Under transmission electron microscope,observation of aortic endothelial cells shows:endothelial cells in direct-moxibustion group,herbal-cake-separated-moxibustion group and herb group are intacter,most of them are normal in forms,internal elastic membrane is continuous,thickness is even,the cells of muscles of medial membrane are relatively normal and the structure of aortic endothelial cells of those groups are similar to those of blank group.But the structure of aortic endothelial cells in model group is different, endothelial cells disappeared,in cytoplasm the sedimentation of a great quantity of lipide can be seen,internal elastic membrane is interrupted,thickness is uneven,there is focal dissolution,the cells of smooth muscle in the medial membrane has sedimentation of lipide,and has frothy change.Conclusion:①Satisfied accommodation on metabolism of lipid,lipoprotein,correlation factor of vascular endothelial cells injury,the expressin of vascular cell adhesion factor of HLP and AS rabbits appeared after treated by direct-moxibustion group,herbal-cake-separated moxibustion and herb group.②In regulating effect on TC of HLP and AS rabbits,each of herbal-cake-separated moxibustion group and herb group is better than direct moxibustion group.③Protective effect to the aortic endothelial cells of HLP and AS rabbits appeared after herbal-cake-separated moxibustion...
