Objective:The project background is discovering 7 specific metablolites as cholic acid from the blood of peptic ulcer patients of qi deficiency and blood stasis syndrome, through metabolomics research approaches and technology. Our aim is to continue the preliminary study, to further verification the existence of the seven specific metablolites as cholic acid, exploring how they are changing between before and after the therapy, by expanding sample capacity. We hope to pave the way for the follow-up study of finding out potential biomarkers for peptic ulcer of qi deficiency and blood stasis syndrome, to provide a theoretical basis of diagnosis, prognosis, and prevention of the disease, and to provide a basis of clinical trials for objective indicators on TCM types of syndromes.Methods:This research sets treatment group and healthy volunteers group, each contains 30 cases, who are all from Suzhou traditional Chinese medicine hospital. The treatment group takes electronic gastroscopy examination, mucosal biopsies, HP test, in addition to give the grading of symptoms and quantification scores, before therapy. While the healthy volunteers group only take the specific metabolites tests. Therapeutic schedule:To take YiQiHuoXue decoction combined with Omeprazole orally, for eight weeks as a treatment cycle. The treatment group review electronic gastroscopy, and also give the grading of symptoms and quantification scores, blood biochemistry analysis and specific metabolites tests, after therapy.Results:Through metabonomics technology, we discovered six special substance, the cholic acid as the early study. The treatment group had lower cholic acid level before therapy, which had distinct difference compare to the healthy volunteers group. But the cholic acid level tend to be normal after therapy, there had no visible difference between two groups. The symptom grading and quantification scores of patients enrolled had distinct decrease after therapy than before, the difference had statistic signification(P<0.05).There had no distinct difference in the TBil level between before and after therapy.Conclusion:The content of the specific metablolites as the cholic acid, in the peptic ulcer patients belonging to qi deficiency and blood stasis syndrome, had regular variations with the change of pathogenic condition, between before and after therapy. The cholic acid may be has some correlation with the peptic ulcer patients belong to qi deficiency and blood stasis syndrome.
