OBJECTIVE To investigate the expression of ErbB2 and Ebpl in gastric carcinoma and to clarify the association between ErbB2 and Ebpl expression and the clinical significance of this expression in patients with gastric carcinoma.METHODS A total of 132 gastric cancer tissues were collected from patients with gastric cancer who underwent total or subtotal gastrectomy at the Yanbian University Hospital from January 2011 to November 2013, and corresponding clinicopathologic parameters wre gathered. Among the 132 patients with gastric cancer,97 cases are male, and 35 cases are female; 85 cases are Han, and 47 cases are Koreans. The patients ranged in age from 38 to 81 years old, the average age was 63.03±9.06 years old, and 42 cases were less than 60 years old,90 cases reached or exceeded 60 years old. The histological type of all the samples were identified as adenocarcinoma after riviewed by two experienced pathologists. According to tumor grade,49 cases were classified as well-and moderately differentiated types and 83 as poorly differentiated type. According to Borrmann’s classification,3 cases were classified as type Ⅰ,101 as type Ⅱ,26 as type Ⅲ, and 2 as type Ⅳ. According to tummor position,13 cases located in Upper,38 in Middle, and 81 in Down. According to the largest diameter of tummor,77 cases were classified as<5cm group, and 55 as>5cm group. In the 132 samples we examined,79 cases were classified as postive for lymph node metastasis, and 53 as negative; 14 cases were classified as postive for metastasis, and 118 as negative. The depth of tumor invasion was determined according to the secenth edition of "The Stomach Cancer TNM. Staging Standard", promulgated by the American Joint Committee of Cancer (AJCC). In the 132 samples we examined,21 cases were classified as T1/T2, and 111 as T3/T4. All the cases didn’t have chemotherapy or radiotherapy before operation. Immunohistochemistry was performed on 132 gastric cancer tissues to detect the protein expression of ErbB2 and Ebpl.RESULTS The overexpression of ErbB2 protein was observed in 38.64% of all the samples, and the rate of the overexpression of Ebpl protein in gastric cancer was 25.76%. Statistical analysis showed that the overexpression of ErbB2 protein was significantly associcated with Borrmann’s classification, tumor grade, the depth of tumor invasion, lymph node metastasis, and metastasis(P<0.05). However no association was shown between the overexpression of ErbB2 protein and sex, age, nation, tumor size, tumor position (P>0.05). The overexpression of Ebpl protein was significantly associcated with Borrmann’s classification, tumor grade, lymph node metastasis, and metastasis (P<0.05), but no association was shown between the overexpression of Ebpl protein and sex, age, nation, the depth of tumor invasion, tumor size, tumor position (P>0.05). The expression of ErbB2 and Ebpl was postively related in gastric cancer (P<0.05).CONCLUSION The combined detection of ErbB2 and Ebpl in gastric cancer is conducive to the judgment of the malignant degree, invasive and metastatic tendency of gastric cancer. The expression of ErbB2 and Ebpl were postively related in gastric cancer tissue, they may have synergistic effect on the promotion of the occurrence and development of gastric cancer, Ebpl may be recognized as a assistant therapeutic target of ErbB2-targeted therapy for gastric cancer.
