This study was designed to investigate the three kinds of Rho A, Snail, VEGF m RNA and protein in Salivary adenoid cystic carcinoma(SACC) tissues and adjacent tissues(control group) in the expression and clinical significance. It discussed the expression of Rho A, Snail, VEGF m RNA and protein in SACC tissues and the relationship with clinicopathological characteristics of the patients. It explored the correlation of Rho A, Snail, VEGF m RNA and protein in different tissue types, TNM stage, lymph node metastasis expression and further explored the mechanism of Rho A / ROCK / PKD1 / NF-κB / Snail molecular signaling pathways of bloody transfer to provide a theoretical basis and therapeutic targets for the SACC invasion and metastasis.We selected 55 cases of complete tissue paraffin blocks(SACC) of Department of Pathology and pathological in September 1997 to September 2013, the First Affiliated Hospital of Hebei North University which consisted of 20 cases from adjacent tissue cancer, aged form 17 to 80 years, with an average age of 48.36 years. The pathological type of salivary gland tumor according to WHO(2005): 20 cases of sieve-like organization, 19 cases of a tubular-type organization and 16 cases of physical organization; we conducted a clinical Staging according to TNM staging(UICC): 37 cases ofⅠ+Ⅱand 18 cases of Ⅲ+Ⅳ. We used HE staining, immunohistochemistry(IHC) and in situ hybridization(ISH) technique to detect the expression methords of Rho A, Snail, VEGFm RNA and protein in SACC and cancer tissues and colleced statistical characteristics of clinical pathological parameters. We analyzed the relationship between index expression and gender, age, size of organization and expression indexes different form tissue types, TNM stage and lymph node metastas is. We applied of Statistical Methods SPSS17.0. Chi-square test was applied between the two groups, Spearman rank correlation test was used between the data and the results obtained with P <0.05 was considered significant.Rho A, Snail, VEGFm RNA and protein was expressed in salivary adenoid cystic carcinoma and adjacent tissues in which positive material of Rho A mainly located in epithelial cell membrane of tumor cells, part of the expression in the cytoplasm and linear brown. The positive material of Snail mainly located in the nucleus of cancer cells and brownish yellow or brown granules. VEGF-positive substance was mainly located in the cytoplasm of tumor cells, brown or tan partic les. The positive expression rates of Rho A, Snail, VEGFm RNA and protein in 55 cases of SACC tissues were as follows: 65.4%(36/55), 63.6%(35/55), 67.3%(37/55) and 69.1%(38/55), 72.7%(40/55), 76.4%(42/55), respectively. The positive expression rates of Rho A, Snail, VEGFm RNA and protein in 20 cases of adjacent tissue expression rates were 5%(1/20), 5%(1/20), 10%( 2/20) and 5%(1/20), 10%(2/20), 15%(3/20), respectively. SACC expression in three groups were higher than the adjacent group, the difference was significant(P<0.05).The high expression of Rho A, Snail, VEGFm RNA and protein in SACC tissues has nothing to do(P> 0.05) with the patient’s sex, age, size and organization Volume. The expression of Rho A, Snailm RNA and protein was significant with its tissue type, tumor grade and lymph node distant metastasis(P <0.05). VEGFm RNA and protein expression was not significant in tissue type, histological grade, lymph nodes distant metastasis and type of organization(P> 0.05). but significant with tumor grade, lymph node distant metastasis(P <0.05); In 55 cases of adenoid cystic carcinoma, the number of cases of positive expression of genes and proteins of Rho A, Snail, VEGF was: Rho A gene was 36 cases, Snail gene was 35 cases, VEGF gene was 37 cases, Rho A protein was 38 cases, Snail protein was 40 cases and VEGF protein of 42 cases. The data we got by spearman correlation analysis and the results showed: Rho A, Snail, VEGF gene and protein expression was positively correlated. Statistics showed that: Rho A, Snail and VEGF expression in SACC was significantly increased compared with adjacent normal tissues, which suggesed that the adenoid cystic carcinoma development, invas ion and metastasis in the process and they might work together to promote mutual partic ipation, and three biological indicators may become a co mmon occurrence SACC invas ion and metastasis, and may contribute to the SACC invasion and metastasis by Rho A / ROCK / PKD1 / NF-κB / Snail signaling pathway joint action provided the treatment of SACC a new theory and therapeutic targets.
