The Effects Of Huaao Capsule On Inflammatory Factors, ICAM-1, VCAM-1 And MCP-1 Of Diabetes With Early Macroangiopathy: An Experimental Study

Objective: T o observe the effects of the Huaao capsule on rat models of diabetes with early macroangiopathy, the weight, blood lipids, blood sugar, inflammatory factor and adhesion molecules were detected for investigating its protective effects on diabetic blood vessels.Methods: The healthy male SD rats, experimental object, were randomly divided into control group and blank module. High sugar, high fat feed were given and STZ were given by intraperitoneal injection to control groups to build module of diabetes with early macroangiopathy rats. The rats that turned into diabetes with early macroangiopathy successfully were randomly divided into model group, the Huaao capsule low-dose group, Huaao capsule high-dose group and Tongxinluo capsule group. The treatment group rats to were treated with huaao capsule suspension and tongxinluo capsule suspension lavage by intraperitoneal injection for 8weeks respectively, while the model group and blank control group were given to the same amount of saline lavage. After that the rats were drugged by general anesthesia, and the abdominal aorta and thoracic aorta were separated. The abdominal aorta blood and thoracic aorta tissue were used to test blood glucose, total cholesterol, triglyceride, TNF-a, CRP, IL-6, ICAM-1, VCAM-1-and MCP-1. In addition, the inflammatory factors were determined by Radioimmunoassay and the adhesion molecule by the method of ELISA(competition).Results:(1) Normal group rats activities freely and were in good condition. While the model group rats appeared polyphagia, polyuria, Polydipsia, weight loss, fatigue, tired, and tail fester undesirable healing, the symptoms of treatment group rats compared with model group were alleviation.(2) In the aspect of rats weight, there was no statistically significant difference compared with prior treatment(P > 0.05); After treatment, model group, Huaao low dose group, Huaao high dose group, Tongxinluo group were compared with the blank control group and the difference was statistically significant(P < 0.05); the weight of Huaao high-dose group was decreased compared with Huaao capsule low-dose group, model groupand and tongxinluo group the difference was statistically significant(P < 0.05).(3)The blood sugar of model groups after modeling for 1 week, 2 weeks and 4 weeks and the treatment group after treated for 8 weeks were deceased compared with blank control group and the difference was statistically significant(P < 0.05); The blood sugar of Huaao high dose group, model group, Huaao low-dose group, Tongxinluo group had no differences for each other(P > 0.05).(4) After treatment, the CHOL, TG of Huaao high-dose group, the Huaao low dose group and Tongxinluo group were lower than that of model group, and Huaao high dose group, Huaao low dose group were compared to model group and the difference was statistically significant(P < 0.05); The CHOL, TG of Huaao high dose group were lower than Huaao low dose group and Tongxinluo group(P < 0.05).(5) The TNF-a, CRP and IL-6 levels of model group compared with blank group were higher significantly(P < 0.05); The differences of the TNF-a, CRP and IL-6 levels between treatment group and model group were of statistically significant(P < 0.05); Furthermore, the experiment results of CRP and IL-6 of Huaao high dose group were lower than Huaao low dose group and Tongxinluo group(P < 0.05); About the lever of TNF-a, Huaao high dose group was decreased compared with other treatment(P < 0.05), but Huaao low dose group(P > 0.05).(6) The ICAM-1, VCAM-1 and MCP-1of model group compared with blank group had higher expression level significantly(P < 0.05), and the treatment group was lower than model group(P < 0.05); The lever of ICAM-1, VCAM-1 and MCP-1 of Huaao high dose group were lower than Huaao low dose group(P < 0.05), but in the aspect of MCP-1 lever there was no statistically difference between Huaao high dose and Tongxinluo group(P > 0.05).(7) Compared with the blank control group, the pathology of the thoracic aortic of model group were different. For the latter, the structure was disorder, with aortic intima thickening, foam cells and lipid deposition gathered in subendothelial tissue, and Interstitial collagen fibers hyperplasia observed in mesolamella. Pathological performance in each treatment group were improved compared with model group, especially the Huaao high dose and Tongxinluo group.Conclusion:(1) T he Huaao capsule has the functions of regulating lipid metabolism, dropping the levels of TG and CHOL, but has inefficacy of lowering blood glucose.(2) The Huaao capsule can reduce the levels of TNF-a, CR, IL-6 and the expression of ICAM-1, VCAM-1, MCP-1derived from thoracic aorta tissue of diabetic vasculopathy model rats, suggesting that Huaao capsule has a protective effects of vessels, whose mechanism may be concerned with the anti-inflammatory mechanism.
