Objective:.To investigate the expression level of CD47 protein in laryngeal squamous cell carcinoma and its impact on the development of the carcinoma and its clinical signifance,and explore the effect of silencing CD47 expression and anti-CD47 antibodies(anti-CD47mAbs) on the biological behaviors of laryngeal carcinoma cells Hep-2.Method: 1.The specimens were divided into laryngeal squamous cell carcinoma group and normal laryngeal tissues group(control group).Western blot were used to detect the expression of CD47 protein.The relationship between CD47 expression level and its clinicapathological parameters was analyzed by SPSS 17.0.2.CD47-RNAi-LV(CD47-RNAi-lentivirus)was transfected into Hep-2 cells. Western blot was used to detect the expression of CD47 protein of the Hep-2 cells pre- and post-RNAi,and chose the most effective RNAi. 3.Compare the influence of CD47-RNAi-LV and the anti-CD47 mAbs on the cell proliferation,apoptosis and invasion ability by MTT、FCM and Transwell invasion assay respectively.Result: The CD47 protein expression in laryngeal squamous cell carcinoma and its normal counterparts were statistically different(p<0.05).The CD47 expression had relationship with the clinical stage and lymph node metastasis while not the age and the degree of differentiation.Expression of CD47 protein was downregulated compared with the negative group by CD47 RNAi.Compared to normal control group,negative control group,MTT showed that from the fourth day on the RNAi group and anti-CD47 mAbs group have the similar RI(Rate of inhibition)(p<0.05),there was no statistical significance between the RNAi group and anti-CD47 mAbs group. The cell apoptosis assay showed that the apoptosis rate of the expremental groups were higher than the control groups.The Transwell invasion assay results showed that with RNAi and anti-CD47 mAbs the number of Hep-2 cell penetrating significantly reduced,lower than the control groups.Conclusion: The high expression level of CD47 in laryngeal cancer suggest that CD47 involved in the process of the tumor development.Silencing of CD47 andanti-CD47 mAbs attenuate the malignant phenotype of Hep-2 cells.
