Objective:To inweste the effect of cetate Alarelin in different does group on the growth of hum an endometrial carcinoma xenograft tumor and expression of DEDD.Methods:Select 4-6 week year old female nude mice inoculated subcutaneously into nude mice near the left forelimb armpit of in vitro cultured endometrial cancer,the establishment of subcutaneous xenograft model. After establishing a successful model, remove a tumor and a tumor nodule largest nodule minimal.Then the remaining 32 mice were randomized into four groups(each eight mice):control group(saline), Alarelin low(20μg/kg),medium(40μg/kg),high(80μg/kg).Continuous observation, Alarelin group was given 0.2ml/d,the control group received saline 0.2ml/d. Measured every four days during a length diameter tumor nodules, and recorded on a growth curve.After four weeks mice were killed and complete removal of the tumor nodules in the left forelimb,measure the volume of tumor nodules.Inhibition rate was calculated for each Alarelin group.The nodules removed to make paraffin sections, HE staining,light microscope morphological changes in tumor nodules cells.Detect the expression of DEDD in the immunohistochemical of tumor tissue.Results:1.With the increase of Alarelin dose,tumor growth curves slope downward trend, the growth rate gradually slowed down,and there was significant difference(P <0.05).2.Tumor volume of Alarelin was significantly smaller than the control group [(898.96± 80.326) than(1707.74 ± 189.918) mm3,P <0.001],the inhibition ratewas 35.43 %.3.HE staining showed that tumor cells arranged in dense,with clear atypia,large nuclei,stained emitotic more,less interstitial in control group.Compared with control group,tumor cell morphology,cell arrangement sparse,showing large unstructured necrosis,tumor cell nuclei stained pyknotic more mitotic less in different dose group.4.Results of Immunohistochemical showed that in the gray values of DEDD protein expression in low,medium and high dose group were 156.2±5.101,142.85±7.266 and113.65±4.381 respectively,they were significantly lower than the control(171.75±8.213,both P<0.05).Conclusions:1.Alarelin can inhibit the growth of human endometrial carcinoma xenograft tumor and with the increasing of Alarelin dose,the inhibition rate increased.2.Alarelin may increase DEDD proteinon on human endometrial carcinoma xenografts in nude mice.
