Object: To investigate the expression of type II 11β-hydroxysteroid dehydrogenase in the lung tissue of neonatal rats. To study the roles of 11β-HSD2 and its signaling factors in pathogenesis of persistent pulmonary hypertension of newborn, and learn more about pathogenesis of Persistent pulmonary hypertension of the newborn(PPHN).Methods: Intrauterine hypoxia and intraperitoneal injection of indomethacin induced rat PPHN model were established. Sprague-Dawley rats on the 19 th day of pregnancy were randomly divided into PPHN and control groups. The PPHN group was intraperitoneally injected with indomethacin(0.5mg/kg) twice daily and exposed in 12% oxygen for three days. The control group was intraperitoneally injected with an equal volume of normal saline and exposed to air. Neonatal rats were born by caesarean section from both groups on the 22 nd day of pregnancy. The expression of 11β-HSD2 in the lung tissue of neonatal rats were observed by Confocal laser technology, and the m RNA levels of 11β-HSD2, SP1 and CREB in the lung of PPHN group and control group were measured by RT-PCR. In addition, serum cortisol levels and prostacyclin, renin, angiotensin II and aldosterone in the lung tissue of both groups were measured using ELISA.Results:(1) The thickness of the right ventricular free were higher than that in control group. The thickness of the right ventricular free And the ratio of media/(media + lumen) thickness was higher than that in control group.(2) The protein of 11β-HSD2 was mainly positive in the cytoplasm of the alveolar epithelial cells, bronchiolar epithelial cells and small pulmonary vascular endothelial cells.(3) The m RNA expression levels of 11β-HSD2, SP1 and CREB in the lung tissue were decreased than that in control group, while serum cortisol levels and renin, angiotensin II and aldosterone in the lung tissue were higher in the PPHN group than in the control group.Conclusion: 11β-HSD2 and its signaling factors play roles in pathogenesis of PPHN.
