Objective:Osteoarthritis is a kind of chronic degenerative disease pathological change in this disease are consist of the destroy of integrality of the joint disease, its pathogenesis is not clear until now. In recent years, with the development of molecular biology, more and more domestic and foreign scholars are beginning to research the relationship of matrix metalloproteinase and OA. This experiment was used to study the MMP-3/-13 expression and correlation in different stages of OA and normal synovium, to elucidate the significance of MMP-3/-13 in the pathogenesis of OA and provide a theoretical basis to find effective interventions target for the clinical treatment of OA.Methods:Eighty cases synoviums are from the patients in department of orthopedics of the first affiliated hospital of Shi He-zi University from the February 2011 to the August 2012,which operation include the knee joint of arthroscopy, arthroplasty, amputation and so on. All of the patients were diagnosed by the doctor of orthopedics or using the proposed criteria from American in1995.The collected synovium were washed three times with saline in one hour after operation, then cut by half synovium into liquid nitrogen, the other fixed by 10% paraformaldehyde. Finally, Immunohistochemically and Western blot were used to detect the expression of MMP-3/-13 protein in eighty cases of osteoarthritis. The results according to SPSS 17.0statistical analysis software: All of the categorical data is signed χ2-test, and the mean will adopt mean 士standard deviation, and the mean is different, two groups compared with T-test, use correlation coefficient analysis between MMP-3and MMP-13,according to P<0.05 presents statistics different.Results:The expression of MMP-3 and MMP-13 protein in osteoarthritis patients synovium of knee joint was markedly higher that in the normal synovium(χ2=29.70/21.99,P<0.01), and the rownish yellow was mainly located in the cytoplasm. In OA group, Pearson correlation coefficient between MMP-3 and MMP-13 is 0.845(r=0.845,P<0.01), the expression of MMP-3 were positively correlated with MMP-13 in OA synovium tissue. Western blot results showed that expression of MMP-3/-13 protein in osteoarthritis patients synovium of knee join(1.147 士 0.219/1.051 士 0.248,P<0.01)was significantly higher than that in the non-osteoarthritis(0.275 士 0.134/0.256 士 0.05,P<0.01), Pearson correlation coefficient between MMP-3 /-13 is 0.997(r=0.997,P<0.01)and were positively correlated with MMP-3 /-13 protein and OA.Conclusion:In OA group, the expression of MMP-3 and MMP-13 protein in synovium tissue were higher than normal group, they were positively correlated with each other and at the same time were close relationship with OA. So they maybe the marker of diagnosis 、judgment of osteoarthritis.
