Part 1 The different expression level of Talin-1 in different prostate cancer cellsand its relationship with cell′s invasive abilityObjective: By culturing different people prostate cancer cells and normal prostate cancer cell lines in vitro,simulated different histological subtypes of primaryprostate cancer and normal prostate model, detect Talin expression in differentcells and compared the invasion and migration ability of different cells,discussthe relationship between invasion and migration of human prostate cancer celland Talin, thus for make a further research for the possible mechanisms ofTalin invasion and migration of prostate cancer and to provide theoretical basis.Methods: Cultured different prostate cancer cells(PC- 3, LNCa P, DU- 145 cell line) and normal prostate cancer cell lines in vitro, subculture to logarithmic phase, using rt-pcr technique to detect the Talin- 1 m RNA expression level of these cell lines,used Western blot technique to detect the Talin- 1 protein expression level of different cell lines,used Transwell invasion and migrationexperiment and cell scratch test to detect the invasion and migration ability ofeach cell line,used MTT method to detect cell proliferation.Result:Compared with the normal prostate cancer cell, Talin- 1 m RNA andprotein expression level of LNCa P cell lines,(PC)- 3 cell line, prostate DU145 cell line with it, P < 0.05;Talin- 1 m RNA and protein expression level wasdecreasing among DU- 145, PC- 3, LNCa P cell lines, comparison betweengroups were statistically significant(P >0.05). Compared with normal prostate cancer cell lines, invasion and migration ability of LNCa P cell lines, PC- 3cell lines, DU145 cell lines was significantly higher(P<0.05), the migration,proliferation and invasion ability decline among DU- 145, PC- 3, LNCa P cell lines, the comparison between groups were statistically significant(P>0.05).Conclusion: 1, the different sources of prostate cancer cell have different invasion and migration ability; 2, Different sources of prostate cancer cells have different Talin- 1 the expression level; 3, Talin- 1 is associated with prostatecancer invasion and migration ability; 4, high express Talin-1 prostate cancer correlate with low invasion and migration ability and low grade, low expressionTalin-1 prostate cancer correlate with strong invasion and migration ability and high malignant degree; 5, Talin-1 can serve as a predictor of prostate cancer.Part 2 the influence of the invasive and expression ability of PC- 3 cell lines after silence Talin- 1 geneObjective: Silenced the abnormal expression Talin- 1 gene in prostate cancerPC-3 cells,study the change of invasion and migration ability of prostate cancer cells after Talin- 1 gene silence, preliminarily discuss using Talin- 1 as the feasibility target genes for specific interference.Methods:Using the method of RNAi,set talin gene as the therapeutic targets for prostate cancer PC-3cells talin gene silencing, and set blank control group,PC-3gene silencing group, gene silence join transforming growth factor(TGF- β) group, PC-3 join TGF-β group, used western blot technique to detect the E- cadherin and Vimentin protein expression levels, used transwell invasionand migration experiment and cell scratch test to detect the invasion and migration ability, used MTT method to detect cell proliferation.Result:After gene silencing, E- cadherin protein expression level of PC-3+ TGF- β, PC-3, PC-3+ TGF-β + RNAi, PC-3 + RNAi groups was increasing order, Vimentin protein expression level and invasion,proliferation and migrationability was decreasing order, differences between four groups were statisticallysignificant(P<0.05).Conclusion: prostate cancer cells growth has slowed after silence Talin- 1 gene, and migration ability of the tumor slow too, Its possible mechanism is topromotethe expression of E- cadherin protein and reduce the expression of Vimentin protein and to induce EMT phenotype transformation and implementation,this test may provide theoretical basis for the prostate cancer Talin- 1 gene therapy.
