Objective:To establish and improve the quality standard of Solanum Lyraturm Thunb, Provide a theoretical basis for the control the quality and safety of clinical medicine of Solanum Lyraturm Thunb.Methods:1 in order to identify its true or false we perform an experiment for morphological identification, microscopic identification and physical and chemical identification of Solanum Lyraturm Thunb.The morphological identification can describe for the original plant identification and medicinal material of Solanum Lyraturm Thunb; Using the optical microscopy for roots, stems, rootstock, powder microscopic identification of Solanum Lyraturm Thunb; Using the TLC distinguish the different batches of Solanum Lyraturm Thunb. 2 According to "Chinese Pharmacopoeia"(2010 edition) Appendix methods for examining the water content, total ashes and acid-insoluble ashes content of Solanum Lyraturm Thunb. 3 We use solasodine as the control for measuring the absorbability of the Solanum lyratum Thumb in the point of 412 nm by the ultraviolet-visible spectrophotometry after is dyed with the bromocresol green and then calculate total solasodine in Solanum lyratum Thumb. 4 According to "Chinese Pharmacopoeia"(2010 edition) Appendix method to determine the content of Pb、Cd、As、Hg、Cu for ensuring the safety of clinical medicine. The content of Pb and Cd was measured by graphite furnace atomic absorption spectrometr, the content of As was measured by atomic fluorescence spectrometry,the content of Hg was measured by cold vapor absorption method, the content of Cu was measured by flame atomic absorption spectrometry. 5 For comprehensive evaluating of the quality of Solanum Lyraturm Thunb to establishing HPLC fingerprint.. Column: waters C18(150.mm×4.6.mm, 5.μm) column(pre-column unprotected live); mobile phase :acetonitrile-0.2% formic acid, gradient elution; flow rate : 1.0 mL.min-1;detection wavelength: 285 nm; column temperature : room temperature.Results:1 the morphological and microsope character of Solanum Lyraturm Thunb is evident; Using cyclohexane: ethyl acetate(2: 1) as the solvent for expanding,the result is that spot of TLC is clear and the effect of expanding is good; 2 according to the result of the water, total ashes, acid-insoluble ashes we draw up the water content is no more than 9.0 percent,total ashes is no more than 10.8 percent, acid-insoluble ashes is no more than. 3 Making use of the ultraviolet-visible spectrophotometry to determine total alkaloids content of Solanum lyratum Thumb, the result of recoveries did not achievement the experimental criteria of 95%-105%,determination of the total alkaloid has not been included in the criteria.. 4 We find that the content heavy metal have difference in different regions through determination of the residues of Solanum Lyraturm Thunb Pb, Cd, As, Hg, Cu by atomic absorption spectrometry. 5 Common pattern of the fingerprints were established, intuitive analysis of HPLC fingerprint from different areas of Solanum lyratum Thunh,and evaluated their similarities through Traditional Chinese medicine(TCM) chromatographic fingerprint similarity evaluation software,compared them and determined 13 peaks in the spectrum as Solanum lyratum Thunh’s characteristic peak.Conclusion:Through analysizing the result of Solanum Lyraturm Thunb from different origin,the method of experiment is reliability and high sensitivity. Establishing fingerprint and combining morphological identification、the content determination which can be integrated quality evaluation of Solanum Lyraturm Thunb and providing the basis for the scientific use of it.
