Objective:Irritable bowel syndrome(IBS) is the most commonly functional disease in clinic. Up to now, its etiology and pathophysiology is still unknown. In recent years, visceral hypersensitivity is recognized as an important pathophysiological mechanism in IBS. In addition, IBS symptoms can be induced or exaggerated by psychosocial stress or acute gastrointestinal infection via Central or peripheral mechanism.It’s reported that the IBS incidence rate in European and American area is 10%-20%, and it’s around 5% to 12% in China. However, its etiology is complex. For many years, the development of IBS research seems to be slow and disappointing. As a reason, IBS research is lacking of advanced animal model, and the purpose of this experiment is to use a combination of Chinese and western medicine to build up a new type of animal model with the purpose of future clinical service.Regarding IBS treatment, western medicine curative effect was not satisfactory in the past. Anyway, the traditional Chinese medicine in the treatment of IBS had profound historical and obvious effect, but the reason was not clear so far.Methods:This experiment is using the combination of Chinese and western medicine, making new IBS rat model that can perfectly simulate the etiology and pathogenesis of IBS symptoms of human diarrhea type.This study is taking TongXieYaoFang as main medicine aiming at treatment of IBS rat model of diarrhea type, targeting at HE staining of rats’brain and intestine, focusing on immunohistochemical detection of the hippocampus of emotional centers of the brain.Results:The new type of animal model based on combination of Chinese and western medicine can be widely used in clinical research.TongXieYaoFang is able to reduce hippocampal FOS expression, which is indicating that TongXieYaoFang plays a regulatory role on the central FOS changes.Conclusion:The new type of IBS rat model based on combination of Chinese and western medicine, can perfectly simulate disease cause and symptoms of human diarrhea IBS type, by having good repeatability and even lasting for months, which can be used in the basic research of traditional Chinese medicine and western medicine.TongXieYaoFang plays a regulatory role on central FOS changes.
