Laryngeal cancer is a common malignancy in otolaryngology-Head and Neck Surgery, which is most common laryngeal squamous cell carcinoma,accumulating statistic data shows that the incidence rate is increasing stablely with each year. The pathogenesis of laryngeal cancer is complex, associated with many different factors, such as genetic background, bad living habits and environmental factors like long-term smoking and drinking, serious air pollution and harmful chemical stimulation, long-term exposure to radioactive nuclide, virus infection, and lack of trace elements and so on. In recent years,study found that the microenvironment in the tumor tissue, such as hypoxia,low sugar, could induce the synthesis of glucose regulatory protein 78(GRP 78)may also ccounts. The expression of GRP 78 increased distinctly in a wide variety of tumors(e.g. vulvar squamous cell carcinoma, esophageal squamous cell carcinoma, breast cancer, etc.). In addition, the expression of calreticulin(CRT) in tumor cells, apoptotic cells, certain drugs, such as anthracyclinebased stimulate, is also higher than normal cells, the cells on the surface of the CRT can combine with immune cell receptors like CD91/l RP1, thus enhancing the immune response. However, the relationship between laryngeal cancer and the abnormal expression of GRP 78 and CRT is rarely studied and far from clear.Objective: Investigate the relationship between the biological behavior of GRP78 and CRT in squamous cell carcinoma of larynx, thus provides new ideas and approaches for the diagnosis and treatment of laryngeal carcinoma.Methods: 40 cases of laryngeal cancer patients were selected from the first hospital, Hebei North University, with both the clinical and pathological diagnosis of laryngeal squamous cell carcinoma(LSCC). Collect both thetissue adjacent to the cancer and laryngeal cancer tissue, and determine the expression level of GRP 78 and CRT with western blot and IHC. Meanwhile,40 cases of vocal cord leukoplakia comparison were selected as control, with which statistical analysis was conducted to determine the correlation of the clinical stage and the expression level of GRP and CRT.Results:1 The expression level of GRP 78 in the laryngeal cancer tissue was significantly higher than in the vocal cords leukoplakia and carcinoma tissue adjacent mucosa(x2=17.894,P<0.05;x2=51.767,P<0.05). And the expression level increases with the clinical stage of the carcinoma tissue and the lymph node metastasis situation.2 Immunohistochemical result shows that CRT expression level is apparently higher in carcinoma tissues than the vocal cords leukoplakia and adjacent tissues(x2=20.337,P<0.05;x2=50.622,P<0.05). With the increase of tumor staging, lymph node metastasis and clinical stage, the expression of the CRT egg white also increased(x2=9.172,P<0.05;x2=14.025,P<0.05).3 According to the correlation analysis, the expression of the CRT 78 and CRT were positively correlated in the carcinoma tissue of laryngeal patients.(R2 = 0.86, P = 0.86).Conclusion: Through this experiment, we can know that GRP 78 and CRT expression level is closely related to the development of laryngeal cancer,and both the clinical pathologic stage was positively associated with laryngeal cancer, our study may potentially provide with a better understanding of the clinical staging and the treatment and prognosis of laryngeal cancer.
