Evodia lenticellata Huang is belongs to the family of Rutaceae, was used as the test material. The main contents of this paper include the following several aspects of research work:(1) The pharmacognosy study of the E.lenticellata Huang;(2) The research about the accumulation of chlorogenic acid, evodin, evodiamin, rutaecarpine and total flavonoids at different parts of E.lenticellata Huang;(3) To optimize the extraction process of total alkaloids and total flavonoids by the method of ultrasonic assisted extraction and response surface. The main research results were as follows:1. Study on the pharmacognosy characteristics of E.lenticellata HuangThe fruit of E.lenticellata Huang is quadrigonal and spheroidicity, the surface is brown and rough with number of punctate protuberance and concave oil drop, star shape crack of four corners in the top of fruit, there are some yellow fuzz in the carpopodium. The fruits are hard and brittle with the strong fragrant smell.The color of medicinal material powder is brown, there are two or three oval cells in the telos of glandular hairs and yellowish-brown inclusion, there are many cluster crystal and square crystal; single grain starch, observed compound grain starch accidentally, the spiravessels and annular vessels were presented under a microscope.2. The content analysis of chlorogenic acid on E.lenticellata Huang.The research is to determine the contents of chlorogenic acid in E.lenticellata Huang in different times by HPLC. The chromatographic separation was carried out using Inertsil ODS-3C18 column(4.6 mm×150 mm,5 μ m), the mobile phase was acetonitrile: 0.2 % phosphoric acid(10︰90), the detection wavelength was set at 327 nm, the temperature was 30℃ and the flow rate was 1.0mL/min. The result showed that there are some difference in the content of chlorogenic acid among different periods, the content of chlorogenic acid were range between 4.69 mg/g to 8.90 mg/g in the leaf, 14.59 mg/g to 27.30 mg/g in the flower and 8.79 mg/g to 16.70 mg/g in the fruit. The accumulation of chlorogenic acid in the same part of different time shows certain regularity.3. The content analysis of evodin, evodiamin and rutaecarpine on E.lenticellata Huang.The research is about the accumulation of evodin, evodiamin and rutaecarpine in the flower and fruit of E.lenticellata Huang. An inertsil ODS-3C18 column(4.6×150mm, 5μm) was used with acetonitrile, tetrahydrofuran and ultra pure water(41︰1︰58) as mobile phase. The flow rate was 1.0 mL/min. The detection wavelength was 225 nm and column temperature was kept at 30℃. The result showed that the content of evodin, evodiamin and rutaecarpine in E.lenticellata Huang increased constantly with the growth of E.lenticellata Huang and reach highest when the fruits were close to maturity,evodin is 12.55 mg/g, evodiamin is 3.40 mg/g and rutaecarpine is 27.39 mg/g.4. The research on alkaloids extraction processTo study the extraction technique conditions of total alkaloids from the fruits of E.lenticellata Huang. The optimum extraction conditions of total alkaloids were obtained as follows: extraction temperature of 50℃, ultrasonic power of 180 W, ratio of liquor to material is 40︰1(mL︰g), ethanol concentration of 70%, ultrasonic time of 40 min and extraction times of one. The extraction rate of total alkaloids reached 12.76mg/g under the optimal extraction conditions. The extraction process obtained by response surface methodology was stable and reasonable,accurate and reliable. It was a feasible method to extract the total alkaloids from the fruits of E.lenticellata Huang.5. The content analysis of total flavonoids on E.lenticellata HuangThe research about the accumulation of total flavonoids in the flower and fruit of E.lenticellata Huang had be made by ultrasonic method. Its quantity is measured by spectrophotography in 500 nm with rutin as reference. The results showed that the content of total flavonoids increased constantly with the growth of E.lenticellata Huang and reach highest when the fruits were close to maturity. The fruits of E.lenticellata Huang are rich in total flavonoids and can be used as a resource to extracte them.6. The research on total flavonoids extraction processTechnology of ultrasonic assisted extracting the total flavonoids from the fruit of E.lenticellata Huang was optimized by using response surface method. The optimum conditions obtained were as follows: ultrasonic power of 240 W, ultrasonic time of 30 min, ultrasonic temperature of 50℃, ethanol concentration of 70%,ratio of liquor to material is 30︰1(mL︰g) and extraction times of one. The extraction rate of total flavonoids reached 220.759mg/g under the optimal extraction conditions.
