Studies On Chemical Constituents And Activities Of CHCl₃ Fraction From "Xue Tong"

Kadsura heteroclita (Roxb) Craib (Schisandraceae) is a plant mainly distributed in the southwest of China. The stems of this plant, called "Xue Tong" by the Tujia minority people, are traditionally used in China to treat rheumatoid arthritis and viral hepatitis. The flavor of it is warm, spicy and sweet, "Xue Tong" is said to promote vital energy and blood circulation, to expel wind-evil and to remove wetness-evil in terms of traditional medicine.The stems of Kadsura heteroclita were isolated and purified by silca gel column chromatography, Sephadex LH-20, MCI and prep-HPLC methods. Our phytochemical investigations of the CHC13 fraction afforded 22 compounds, while 18 compounds chemical structures were identified. Their structures were elucidated by spectroscopic analyses and by the comparison of the NMR data with those reported previously. There were nine triterpenoids and steroids:cycloartenone (1), schisanlactone E (2), heteroclialactone A (3), heteroclialactone H (4), schisanlactone c (5), nigranoic acid (6), changnanic acid (7), β-sitosterol (8), daucosterol (9); six lignans:coumarinlignan (10), meso-dihydroguaiaretic acid (11), dihydroguaiaretic acid (12), isolariciresinol (13), burselignan (14), galgarain (15); three sesquiterpenes:6a,9a-dihydroxyca-dinan-4-en-3-one (16), guai-3-en-10α-ol (17), cryptomeridol (18). Compounds 10,16,17 were the new compounds, while 5,13,14,18 were firstly reported from this plant.Partial compounds were tested for anti-HBV activity. Compound 10 showed good inhibition rate against HBeAg and HBsAg (48% and 57%) at 25 ug/mL,10 also demonstrated morderate activity against HSC-T6. Schisanlactone E (2) and coumarinlignan (10) exhibited neuroprotective effects and antineuroinflammation activities. The antimicrobial activities in vitro were tested for 7 compounds,11 showed anti-microbial activities (IC50 =6.29 μg/mL,18.04 μg/mL).
