Mosla chinensis Maxim is Chinese traditional herb and belongs to Labiatae and Molsa, which is annual herbage. On the basis of experiments and researches, it’s characteristics of growing and developing was revealed through researches on cultivation methods, and the dynamic changes of target compounds was observed by establishing of an accurate GC method. The experiment for Mosla chinensis Maxim, voitale oil extraction by steam distillation technique was carried out in Box-Behnken design. In order to increase the purity and quality of Mosla herb oil, molecular distillation technology was first adopted on the study of Mosla chinensis oil purification, meanwhile, the purification effect by column chromatography was discussed, a comparison of Mosla chinensis oil after refining by these two different purification methods was shown. The main conclusions are as follows:1.With different sowing time treatment, the characteristics of growing and developing of Mosla chinensis Maxim was revealed, the optimum sowing date is from early spring to April 10th. Based on the result of seedling methods comparison experiment, the yield by thinning is higher than transplant. The yield can be efficiently increased plastic-film mulching technique, the yield of black plastic-film mulching and white plastic-film mulching increased 2717.95Kg/ha (31.6%) and 2333.34Kg/ha (27.2%) compared to the non-treatment group, no significant differences is shown in the yield of black and white plastic mulching treatment. The yield rises dramatically when planting in greenhouse, the yield increased 1807.70Kg/ha (22.7%). The planting pattern of flat planting and bed-planting effects the growth of Mosla chinensis Maxim, but no significant difference in the yield. The experiment result of different density shows:the yield reached maximum of 18923.08Kg/ha with planting distance of 8cm and row space of 25cm.2.GC method for identifying the target compounds of Oxyzymol & Thymol was established, so that the content of Oxyzymol & Thymol in different growth periods and various parts of Mosla chinensis Maxim.were traced. It is shown in the result that the content of Oxyzymol & Thymol in the plant of Mosla chinensis Maxim, reached the peak of 2.80% in August, and leafs group contains the highest proportion of 2.92%.3. By Using stream distillation method, the aboveground part of Mosla chinensis was extracted for violate oil. The experiment was carried out in Box-Behnken design. Taking 3 factors of material-water ratio, soaking time and distillation time as independent variables while extraction rate of volatile oil as the dependent variable, a multivariate second degree polynomial model was established. The extraction parameters were optimized as material-water ratio:1:8.45, soaking time 5.43h and distillation time 4.69h, under these conditions, the final extraction rate is (1.98±0.04)% in average.4.Different elution systems were tested by TLC for separating Mosla chinensis raw oil, the elution system of Ehanol/Acetic Acid/Petroleum Ether (1/0.2/9,v/v/v) was selected. The parameters for refining Mosla chinensis raw oil in molecular distillation are as following, system pressure of 1*10"2mbar, rotor speed ranges from 110 r/min to 170r/min, material-feeding rate in lml/min to 2ml/min, distillation temperature ranges from 40℃ to 60 ℃. It is shown in GC result that the purity of Thymol and Oxyzymol after molecular distillation is 38.25% and 50.09% respectively; the purity of Thymol and Oxyzymol in concentration of collection by column chromatography is 30.47% and 32.49% respectively; while the original content in raw oil of Thymol and Oxyzymol is 17.00% and 21.61%. Both purification methods achieved effective refining effect.
