Objective: To study the expression of NFAT signaling pathway and its downstream molecules in peripheral blood T lymphocytes in Xinjiang Kazakh patients with essential hypertension(EH), and observe the effects of telmisartan on the pathway.Methods: The 60 untreated Kazakh hypertensive patients and 30 Kazakh healthy subjects from Xinjiang were selected randomly in this study. The 60 untreated subjects were divided into 2 groups randomly: intervention group and hypertensive group, T- lymphocytes were isolated from peripheral blood with magnetic cell sorting and cultured for 120 h. mRNA and protein’s expression level in 3 groups were detected by the technique of quantitave reverse transcription polymerase chainreaction and western blot a nalysis respectively. Results: No significant differences in baseline data between these three groups were observed(P>0.05). Both mRNA and protein expression of NFATc1, interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) were found to be expressed in the peripheral blood T lymphocytes.(1) Compared to the controls, the mRN A and protein levels of NFATc1, IL-6 and TNF-α were significantly increased in hypertensive group(P<0.05).(2) Compared to the hypertensive group, the mRNA and protein levels of NFATc1, IL-6 and TNF-α in intervention groups were decreased significantly(P<0.05)(3) Compared to the controls, the mRNA levels of NFATc1, IL-6 and TNF-α were increased slightly in intervention group(Except the NFATc1 all P<0.05).(4) Compared to the controls, the protein levels of NFATc1 elavated slightly in hypertensive group,meanwhile, the protein levels of IL-6 and TNF-α were slightly decreased in hypertensive group(P>0.05) Conclusion: NFAT signaling pathways activate and participate in the pathological process of inflammation in essential hypertension and play an important role in Xinjiang Kazakh patients with essential hypertension.The intervention oftelmisartan can reduce the production of inflammatory cytokines in the etiology and development of EH.
