Purpose: The AA animal models were stablished by benzene and cyclophosphamide(CTX). Respectively with low, medium and high dose Zuogui Marrow-blood Generation Granules and stanozolol treatment of AA rats,compared with normal group, model group, Zuogui high,medium and small dose group and stanozolol group in AA rats spleen T- bet, GATA- 3, IFN- gamma changes in protein expression level and the connection between the three, Zuogui Marrow-blood Generation Granules for benzene and treatment mechanism of AA rats induced by cyclophosphamide, provide experimental basis for the study of the theory of the AA treatment, for the treatment of AA, opening up new research ideas.Methods:72 adult healthy SD male rats, adaptive feed after 1 week,randomly grouped into: 12 normal group, model group, 60, the model group was1 mL/kg body weight subcutaneous injection of benzene in 7 weeks,qod;In 39, 40,41, 42, 43 days according to 25 mg/kg intraperitoneal injection of CTX.The model group rats were randomly divided into 5 groups as model group,stanozolol group, small, medium and large dose groups.Group according to the division of stanozolol group 2.8 mg/kg, small dose group 0.35 g/kg, middle dose group 0.7g/kg, and high dose group 1.4 g/kg to fill the stomach, gastric volume of 1ml/l00 g weight, once a day.Model group and normal group rats given amount of deionized water to fill the stomach.4 weeks after treatment, under aseptic conditions, take the rats spleen, Western-blot detection of AA rats spleen IFN-gamma, T- bet, GATA- 3 protein expression.Result:1. The effects of Zuogui Marrow-blood Generation Granules on T-bet:In the normal group and model group rats spleen are there is the expression of T-bet, compared with normal group, model group rats spleen T-bet protein expression in the higher(p<0.01); compared with model group, stanozolol group and high, medium and small dose group of T-bet protein expression weresignificantly decreased(p<0.01). Compared with stanozolol group, high-dose group of T-bet protein expression had no significant difference(p>0.01), medium and small dose group of T-bet protein expression significantly increased(p<0.01).Compared with large dose group, middle and small dose group of T-bet protein expression significantly increased(p<0.01).Compared with the middle dose group,small dose group of T-bet protein expression has no obvious difference(p>0.01)2. The effects of Zuogui Marrow-blood Generation Granules on GATA-3:In the normal group and model group rats spleen are there is the expression of GATA-3, compared with normal group, model group rats spleen GATA-3protein expression in the lower(p<0.01); compared with model group, stanozolol group and high, medium and small dose group of GATA-3 protein expression were significantly increased(p<0.01). Compared with stanozolol group, zuogui group of GATA-3 protein expression had no significant difference(p>0.01).Compared with large dose group, middle and small dose group of GATA-3protein expression significantly decreased(p<0.01).Compared with the middle dose group, small dose group of GATA-3 protein expression has no obvious difference(p>0.01)3. The effects of Zuogui Marrow-blood Generation Granules on IFN-γ:In the normal group and model group rats spleen are there is the expression of IFN-γ,compared with normal group, model group rats spleen IFN-γprotein expression in the higher(p<0.01); compared with model group, stanozolol group and high, medium and small dose group of IFN-γ protein expression were significantly decreased(p<0.01). Compared with stanozolol group, high-dose group of IFN-γ protein expression had no significant difference(p>0.01), medium and small dose group of IFN-γ protein expression significantly increased(p<0.01).Compared with large dose group, middle and small dose group of IFN-γ protein expression significantly increased(p<0.01).Compared with the middle dose group,small dose group of IFN-γ protein expression has no obvious difference(p>0.01)Conclusion:Zuogui Marrow-blood Generation Granules can improve the hematopoietic function of AA rats, adjust the immune mechanism of AA rats,delay the progress of the disease rat AA, possibly through regulation of IFN-gamma/T- bet related signal transduction pathways.
