| Objective:To explore the expression of SPINK5 in cutaneous malignant melanoma paraffin-embeded tissue samples and its status in occurrence and progress of melanoma. In order to find a new therapeutic target of cutaneous malignant melanoma during its development, and provide important clinical basis of pathogenesis and early intervention for the cutaneous malignant melanoma prognosis in Yunnan region.Methods:Using IHC to detect the expression of SPINK5 in cutaneous malignant melanoma and benign skin nevus tissue samples. And analyze the correlation between its expression and clinical parameters in melanoma.Results:·The expression of SPINK5 in cutaneous malignant melanoma is lower than skin nevus, and the difference is significant (p<0.05)It showed that the expression of SPINK5 in 47 cases of cutaneous malignant melanoma tissues had 16 positive expression cases (34.04%) and 31 negative expression cases (65.96%). In 57 skin nevus tissues,39 cases had positive expression (68.42%) and 18 cases with negative expression(31.58%). The difference was statistically significant, (p<0.05).It showed that low SPINK5 expression level may be important factors, which have contributed to the occurrence of cutaneous malignant melanoma· The expression of SPINK5 decreased in advanced cutaneous malignant melanoma, but no significant difference. (p>0.05)Clinical subgroup analysis found that the positive rate of expression SPINK5 gradually decreased with clinical stage, but no significant difference. And there was no significant correlation between the SPINK5 expression level and clinical parameters.· The expression of SPINK5 in metastasis melanoma is higher than primary melanoma, and the difference is significant(P<0.05)Through NCBI database, we found that in 31 cases of primary cutaneous melanoma tissues, SPINK5 expression levels was higher than 52 cases of metastases tissues, (p<0.05). However in our study, the expression of SPINK5 in metastasis melanoma is higher than primary melanoma, and the difference is significant (76.92 vs 26.92,p<0.05).Conclusion:The expression of SPINK5 in cutaneous malignant melanoma tissues was significantly lower than skin nevus, suggesting lower expression SPINK5 may contribute to the occurrence of cutaneous malignant melanoma. |
