Objective: Malignant melanoma(MM) is a kind of malignant tumor which isoriginated in melanocytes with high degree of malignancy.In recent years,many studies suggest that in cell mitosis process,abnormal expression of theproteins play an important role in the occurrence and development process.Toinvestigate the expression of mitosis-related proteins Aurora-A,TPX2andFBXW7by immunohistochemistry in MM parts and compound nevus parts,explore their significance and relationship in MM parts and compound nevusparts.Methods: The collection of17cases of MM patients and20cases ofcompound nevus patients as controls, were confirmed by HE staining for theMM, compound nevus tissue. These specimens were divided into two groups:the MM group and the compound nevus group, and were examined byEliVisionTMplus Immunohistochemistry. To analyze the possible relationshipand significance of Aurora-A、TPX2、FBXW7in MM group and compoundnevus group. The statistics processing was all conducted by SPSS for windowsversion17.0. The comparisons of rates were analyzed by Fisher exactprobability test. While the correlations of both factors were examined by thePearson correlation analysis.Results: 1.The expression of Aurora-A: Aurora-A protein was stain in the cytoplasmby Immunohistochemistry. The positive rates of Aurora-A protein in17casesof exposure parts of MM were76.47%,20cases of compound nevus were10.00%. The positive rate of Aurora-A in MM group was significant higherthan the compound nevus group(P<0.01).2.The expression of TPX2: TPX2was stain in the nucleus by Immunohistochemistry. The positive rates of TPX2protein in17cases of exposure parts of MM were82.35%,20cases ofcompound nevus were5.00%. The positive rate of TPX2in MM group wassignificant higher than the compound nevus group(P<0.01).3.The expressionof FBXW7: FBXW7was stain in the nucleus by Immunohistochemistry. Thepositive rates of FBXW7protein in17cases of exposure parts of MM were29.41%,20cases of compound nevus were85.00%. The positive rate ofFBXW7in MM group was significant lower than the compound nevus group(P<0.01).Conclusion:1. The positive rate of Aurora-A in MM group was significant higher thanthe compound nevus group. The high expression of Aurora-A plays animportant role in the occurrence and development process of MM.2. The positive rate of TPX2in MM group was significant higher than thecompound nevus group. The high expression of TPX2plays an important rolein the occurrence and development process of MM.3. The positive rate of FBXW7in MM group was significant lower than thecompound nevus group. The low expression of FBXW7plays an important rolein the occurrence and development process of MM. 4. There was a significant positive correlation between the expression ofAurora-A and TPX2, There was a negative correlation between the expressionof Aurora-A and FBXW7, there was no correlation between the expression ofTPX2and FBXW7. They all showed abnormal expression in MM, they couldbe play an important role in the development of malignant melanoma of theskin.
