It was one of the most commonly herbs on Angelicae sinensis Radix for clinical treatment, it has good functions of tonifying and promoting blood. At present, it was widely reported on chemical components, resources distribution and pharmacological effects of Angelicae sinensis Radix. However, it were less research of chromatogram-effect relationship on volatile oil from Angelica Sinensis Radix(OVAS) and quantitative research of MIR spectra by polysaccharide. Based on the methods of Traditional Chinese medicine analysisand Chemometrics, the following some systematically researched:Chapter 1. Studies on chromatography fingerprint of OVASOVAS were extracted by steam distillation, gas chromatography-mass spectrometry(GC-MS) was used as analytical tools. It were commonly qualitatively researched using NIST and retention index. The main chemical compositions including long-chain alkane, olefin, monoterpene, sesquiterpenes, aromatic aldehyde and sesquiterpineol, and less fatty acid, it was a total of 35 compounds, such as the content of Z-Ligustilide(68.84%) was the highest, the content of Z-Butylidenephthalide was the second, and there was a small amount of α-pinene and trans-beta-Ocimene, etc. By similarity analysis and cluster analysis(CA), As you can see similarity calculation results that angle cosine methods were higher and closer, and some samples of similarity were equal, among were greater than or equal to 0.75, many similarity were between 0.80 and 1.00; From cluster analysis result, it had certain relations with geographical environment and soil type. The results showed that the origin of OVAS had a larger difference, it had the certain influence of Environmental factors for OVAS.Chapter 2. Correlation Analysis between OVAS and Its Effects on HUVECs ProliferationOur studies showed that it was existed certain differences on promoting HUVECs proliferation from different regions of OVAS. The chromatogram-effect relationship of volatile oil and promoting HUVECs proliferation effect could be established by Linear regression analysis from Minitab 17 Statistical Software, cell proliferation was regarded as dependent variable(Y), the seven common peaks was viewed as independent variable(X1, X2, X3, X4, X5, X6, X7), the seven peaks issuccessively representative:(1R)-α-pinene, trans.-beta.-Ocimene, 2,4,6-Octatriene,2,6-dimethyl(E,Z), 2-Methoxy-4-vinylphenol, 1H-Cycloprop[e]azulen-7-ol-decahydro-1,1,7-trimethyl-4-m, Z-Butylidenephthalide, Z-Ligustilide, Using Linear regression analysis, X1 and X2 peaks with cell proliferation rate were positive correlation, it was indicated that the chemical composition of its representative with the increase of content of chromatographic peak X1 and X2, its promoting cell proliferation effect was weaken. Other peaks were negative correlation, among X3, X6 and X7 peaks were larger than the other of linear regression coefficient, It was showed the greater the effect on promoting cell proliferation with the content increase of its representative chemical component.Chapter 3. The qualitative and quantitative study of MIR polysaccharides on Angelicae sinensis RadixThe qualitative of MIR showed that It were commonly compared researched using original spectrum with the first derivative spectrum, the result was unable to identify directly, because the the similarity was higher and had more overlapping absorption peak. The second derivative infrared spectrum from the different regions on Angelicae sinensis Radix could preferably identified overlapping absorption peak of original spectrum. In the peak of 2360 cm-1 and the region of 1570 cm-1 to1456 cm-1had obviously different from every region from herb.Our researches showed that the percentage content of total polysaccharides was 3.72 ~ 28.84%, Acid polysaccharides was range from 0.035 mg·mL-1 to 0.517 mg·m L-1, Neutral polysaccharides was range between 0.095 mg·m L-1 to 0.766 mg·mL-1. With the established quantitative model of PLS(Partial Least Square) by the percentage content of total polysaccharides, the polysaccharides from Angelicae sinensis could be rapidly determined, the result showed: coefficient(Rc), RMSEC, RMSECP were successively 0.9924, 0.052 and 0.062, The total polysaccharides of average prediction recovery was 103.55% on Validation sample. Thus it can be seen that the established PLS model was relatively ideal.
