Study On The Quality Standard Improvement Of Zhuangyaojianshen Pill

Objective:To mature the quality standard of Zhuangyaojianshen pills, the topic researched on improvement in Zhuangyaojianshen pills original standard and a more reliable TLC identification method / HPLC content determination was established.Methods:(1) Adopted thin layer chromatography(TLC) to identify four kinds of Chinese herbal medicines in the recipe, and established the TLC identification methods of them.(2) Adopted high performance liquid chromatography(HPLC) to determine the contents of Quercetin and Caffeic acid, and established HPLC methods for determination the content of them.Results:(1) Established the TLC differential conditions of Rhizoma cibotii, Caulis Spatholobi,Taxillus and Cherokee rose. the TLC spots were clearly and separated well, and the negative control did not impose any disturbance.(2) A HPLC conditions for determination of quercetin and Caffeic acid content was found: This analysis was carried out on DIONEX Acclaim? 120(4.6mm× 250 mm, 5μm),with the mobile phase was methanol-0.1%phosphonic acid(53:47), whose elution flow rate was 1.0m L/min, the detection wavelength of 370 nm.The theoretical plate number according to the peak of quercetin in calculation of not less than 5000. Also, The analysis of Caffeic acid was carried out on DIONEX Acclaim? 120(4.6mm× 250 mm,5μm), the mobile phase was methanol-0.1%phosphonic acid(20:80)with flow rate of 1.0 m L/min isocratic elution, detection wave length was 323 nm.The theoretical plate number according to the ic acid peak calculation of not less than 4500.Conclusion:The Quality control standard of Zhuangyaojianshen pill could be improved,both the established TLC and HPLC method in topic were reliable and specific. It would be beneficial to the quality control of the production and the further research of Zhuangyaojianshen pill.
