Study On Chemistry And Pharmacodynamics Of Compound Changkangning Capsules

Objectives: The research object of compound Changkangning are Gentianopsis paludosama and Sophora alopecuroides,from the chemical method and pharmacodynamics method to filter out the best compatibility proportion of Gentianopsis paludosa and Sophora alopecuroides To provide experimental basis for the compound compatibility of Changkangning capsules.Methods:In accordance with the provisions of the dosage of Gentianopsis paludosama and Sophora alopecuroides to design the compatibility proportion of the two drugs.By HPLC method,determination of the content of different compatibility proportion of Gentianopsis paludosama of Luteolin, swertianolin and swertisin.By HPCE,determination of different compatibility proportion of Sophora alopecuroides contains sophoridine, matrine and Oxymatrine content,The combination of the two, from the chemical method to screen the best compatibility of Changkangning compound.Through the study of different compatibility proportion of Gentianopsis paludosama and Sophora alopecuroides in analgesic,anti-inflammatory, antidiarrheal efficacy study. From pharmacodynamics methods to screen out the best compatibility proportion of Gentianopsis paludosama and Sophora alopecuroides.Combined with chemical methods and pharmacodynamics methods, selected the best compatibility proportion of Gentianopsis paludosama and Sophora alopecuroides.Results:According to the results of chemical test, when Gentianopsis paludosa constant, with the increase of the dosage of Sophora alopecuroides, Gentianopsis paludosa swertisin,swertianolin and luteolin content also showed obvious increasing trend,however, the clinical application of the dosage of Sophora alopecuroides is not the bigger the better, because Sophora alopecuroides toxic, clinical dosage should not be more than 0.9g/ days.When the Sophora alopecuroides constant, with the increase of the amount of Gentianopsis paludosa,the content of Sophora alopecuroids sophoridine, matrine and Oxymatrine was also significantly increased.So in line with the premise of medicinal dosage standard and preferred Gentianopsis paludosama and Sophora alopecuroides dosage was large for the best compatibility group,and Gentianopsis paludosa: Sophora alopecuroides =20:0.9 as the best compatibility group.According to pharmacodynamics test, in different compatibility proportion of Gentianopsis paludosama and Sophora alopecuroides and increase Gentianopsis paludosa for compatibility extract analgesic, anti-inflammatory, anti diarrhea effect significantly, increase of Sophora alopecuroides on compatibility extract analgesic,anti-inflammatory effects have enhancement effect, but effect of anti diarrhea effect is not significant.Mice on compatibility extract maximum tolerated volume(total dose) for 32.0g/kg.The maximum amount of tolerance of Sophora alopecuroides 1.52g/kg, display Gentianopsis paludosa: Sophora alopecuroides =20:0.9 are more than adults clinically daily doses of 100 times, to the maximum tolerated dose(MTD) experimental animal death of heart, liver,spleen, kidney, detection of lung were not found pathological changes; Gentianopsis paludosa:Sophora alopecuroides =20:0.9 extract oral safe and reliable.Conclusions:As indicated by the above test results, the best ratio of compound Changkangning preparation screening method is reasonable, safe and reliable, Gentianopsis paludosa: Sophora alopecuroides =20:0.9 can compound sausage as Corning preparation of compatibility proportion, compound Changkangning formulation development development provide compatibility of basic research.
