Rectal cancer is one of highly digestive malignant tumor, and it is the diseases of cancer with poor prognosis. At present, there is a lack of accurate tumor markers, and the early diagnosis of the tumor is mainly rely on fecal accult blood tests, the positive patients through colonoscopy to take the pathology. Most patients with rectal cancer have been found to be in advanced stage. The incidence and mortality of rectal cancer are high, and there is a rising trend. The treatment of rectal cancer is mainly based on surgery, adjuvant chemotherapy, to improve the survival and quality life of patients. Patients with rectal cancer at the end often are died of liver metastasis. With the development of molecular immunology and complement system, and more and more scholars’ pay attention to it.Complement regulatory proteins can regulate the complement system activation and suppression in order to protect the normal tissues, and effectively killing foreign and abnormal cells. In recent years, the study found that complement regulatory proteins play an important role in tumor immune escape, and CD46, CD55 and CD59 are related with human tumor. This research mainly is through the method of immunohistochemistry to detect three complement regulatory proteins expression levels in rectal cancer tissues, and explores the role of CD46, CD55 and CD59 in rectal cancer immune escape, and to knowledge the rectal cancer occurrence development mechanism. To analyze the relationship between the three complement regulatory proteins and the clinical pathological characteristics of colorectal cancer, and to provide a new direction for the diagnosis, treatment and prognosis of rectal cancer. Objective:To analysis the expression of complement regulatory proteins CD46, CD55 and CD59 in colorectal carcinoma and adjacent normal tissues is different. To inquiry the complement regulatory proteins CD46, CD55 and CD59 in rectal cancer in the role of immune escape understands the rectal cancer occurrence and development. And maybe it can provide a new direction for the clinical diagnosis and treatment of rectal cancer.Methods:76 cases pathological block of rectal cancer tissues were found which have been by surgical resection and postoperative pathological diagnosis, 76 cases pathological wax block of normal rectal tissues adjacent to carcinoma, using the immunohistochemical of Envision detected the expression level of CD46, CD55 and CD59 in rectal cancer, and by analysising the relation between rectal cancer clinical pathological features and CD46, CD55 and CD59. The data was analyzed by using SPSS 20.0,using the χ2 test to analyze the expression of CD46,CD55 and CD59 in the rectal cancer. And to analyze the relationship of CD46,CD55 and CD59 with the clinicopathologic features of rectal cancer, using the Spearman correlation analysis analyzed the correlation between CD46,CD55 and CD59.It used α = 0.05 as inspection standard, P<0.05 for differences with a statistical significance. Results:The expression of complement regulatory proteins CD46,CD55 and CD59 in rectal cancer tissues and cancer adjacent normal tissues are different. The positive expression is mainly located in the cell membrane and cytoplasm. The positive rates of cancer tissues are 47.9%(44/76),81.6%(62/76) and 81.6%(62/76).The positive rate of normal tissues are 14.5%(11/76),9.2%(7/76) and 14.5%(11/76).The expression of CD46,CD55 and CD59 is significantly higher than that in the adjacent normal tissues(P<0.05).The expression of CD46 in rectal cancer tissues does not exit correlation with age, gender, tumor size, stage of TNM,lymph node metastasis and distant metastasis(P>0.05). The expression of CD55 and CD59 in rectal cancer tissues does not exit correlation with age,gender(P>0.05),but it exits correlation with tumor size, stage of TNM,lymph node metastasis and distant metastasis(P<0.05).Conclusion:The expression of complement regulatory proteins CD46,CD55 and CD59 in rectal cancer is significantly higher than that in adjacent normal tissues. It suggests that thay may be involved in the decelopment process of rectal cancer as important factors. The expression of CD46 in rectal cancer tissues does not exit correlation with age, gender, tumor size, stage of TNM,lymph node metastasis and distant metastasis. The expression of CD55 and CD59 in rectal cancer tissues does not exit correlation with age, gender, but it exits correlation with tumor size, stage of TNM,lymph node metastasis and distant metastasis. And the later the stage of the tumor, the lower the degree of the expression level of CD55 and CD59 of tumor differentiation. CD46,CD55 and CD59 are important factors in complement activation system. CD46,CD55 and CD59 is highly expressed in rectal cancer. They may be play important roles in rectal cancer cells escaping from the membrane attack complex. It may provide new way to the diagnosis and treatment of the rectal cancer by studying the biological behavior of CD46,CD55 and CD59.
