Objective: Investigate the effect of sports anemia on the rat nervous system and theaction of PNS in the sports anemia development and transfer process on rat nervous system.Methods: The24healthy male4-week-old SD rats were randomly divided into Controlgroup (abbreviated as C group), Sports group (abbreviated as S group) and PNS interventiongroup (abbreviated as P group). We drove the S group and P group rats to do increasing loadtreadmill exercise for ten weeks. And from the fifth week, we gave the P group rats PNS drugintervention, and gave the rats of other group equal volume saline. We cut all rats’ tails to gettheir blood24hours after the last training. Then we measured the Hb concentration and RBCcount in the blood. After36hours of the last training, we sacrificed all rats, taken the heartblood and hippocampal. Then we measured the MDA content, SOD and CAT activity in theserum, and measured the Bcl-2mRNA and BDNF mRNA gene expression in hippocampal byreal-time PCR technology, measured Bcl-2and BDNF protein content in hippocampal byELISA technology. The results were statistically analyzed using SPSS16.0software.Results: The Hb concentration of S group was significantly lower than C group (P<0.05),the Hb concentration of P group was significantly higher than S group (P<0.05), the Hbconcentration of P group was no significant difference with C group, the RBC count of Cgroup, S group and P group were no significant difference. The MDA content and CATactivity in serum of S group were significantly higher than C group (P<0.05), the MDAcontent in serum of P group was significantly lower than S group (P<0.05), the CAT activityin serum of P group was significantly higher than S group (P<0.05), the MDA content andCAT activity in serum of P group were no significant difference with C group, the SODactivity in serum of C group, S group and P group were no significant difference. The Bcl-2mRNA gene expression and Bcl-2protein content in hippocampal of S group were significantly lower than C group (P<0.05), the Bcl-2mRNA gene expression and Bcl-2protein content in hippocampal of P group were significantly higher than S group (P<0.05),the Bcl-2mRNA gene expression and Bcl-2protein content in hippocampal of P group wereno significant difference with C group. The BDNF mRNA gene expression and BDNF proteincontent in hippocampal of S group were significantly lower than C group (P<0.05), the BDNFmRNA gene expression in hippocampal of P group were significantly higher than S group(P<0.01), the BDNF protein content in hippocampal of P group were significantly higher thanS group (P<0.05), the BDNF mRNA gene expression and BDNF protein content inhippocampal of P group were no significant difference with C group.Conclusion: Ten weeks increasing load treadmill exercise can strengthen lipidperoxidation, increase free radical production, decrease antioxidative capacity, cause sportsanemia. Sports anemia can cause rat brain occur hypoxic-ischemic injury and significantlyincrease hippocampal neurons apoptosis. PNS can effectively improve the oxidative stressstatus, reduce the level of lipid peroxidation, enhanced the body’s ability to defense freeradical damage. PNS can promote the expression of Bcl-2and BDNF in rat hippocampal,inhibit neuronal apoptosis in rat hippocampal neuron and play a protective role in sportsanemia caused brain hypoxic-ischemic injury.
