Objective:We aim to discuss the effect of HIF-1α on nucleoprotein ERRα, ERRγ and their target genes when give non-transgenic mice, HIF-1α low-expression mice and HIF-1α high-expression mice with 1 and 6 hours of disposable endurance exercise.Methods:Non-transgenic mice (N), HIF-1α low-expression mice (L) and HIF-1α high-expression mice (H) were divided into control group,1 hour of disposable endurance exercising group and 6 hours of disposable endurance exercising group respectively. All the endurance exercising groups are trained treadmill running at 5% degree and the speed of 20 m/min for 1h or 6h.3 hours after the exercising, skeletal muscle was collected. Nucleoprotein ERRα and ERRγ are tested by Western blotting and their target genes MCAD, PPARα and NRF1 mRNA expression tested by Real Time-PCR.Results:(1) ERRα, ERRγ and mRNA of their target gene increase in non-transgenic mice after 1 and 6 hours of disposable endurance exercising. (2) ERRα, ERRγ and mRNA of their target gene increase in HIF-1α low-expression mice while decrease in HIF-1α high-expression mice. (3) After 1 hour of disposable endurance exercising ERRγ and mRNA of its target gene increase in HIF-1α low-expression mice while decrease in HIF-1α high-expression mice. (4) Each of ERRα, ERRγ and mRNA of their target gene expression has no obvious change after 6 hours of disposable endurance exercising.Conclusions:(1) Along with the time extension, the expression of ERRα/γ in mice skeletal muscle augment. (2) The mRNA of MCAD and PPARα increase in HIF-1α low-expression mice and ERRα decrease in HIF-1α high-expression mice significantly. (3) After 1 hour of disposable endurance exercising ERRγ and mRNA of its target gene increase in HIF-1α low-expression mice while decrease in HIF-1α high-expression mice. |
