Increasing load aerobic training rats model, this paper extracts the mitochondria inside the rat quadriceps, determination and energy metabolism related biochemical indicators, representative for the application of curcumin anti-aging provide some experimental basis.Research purposes: based on the determination of curcumin combined with incremental load after aerobic exercise on aging rat skeletal muscle mitochondria inhibits PTP opening degree, mitochondrial respiratory chain complex enzyme complex III, IV activity and the change of ATP enzyme activity, and were observed under electron microscope mitochondria structure changes, analyze each aging rats.Curcumin combined with incremental load aerobics on aging effect of mitochondrial function in rats.Methods: 21 months only 40 male rats, adaptability to rest 3 days, adaptive training for five days SD male rats were randomly divided into 4 groups, respectively is: quiet control group(N), the curcumin group group(I)(C), incremental load increasing, curcumin + load aerobic exercise group(A), each group of 10, only 2 per component cage, each cage 5 only.Groups(I) and increasing load of curcumin + aerobic exercise group(A) increasing load using the treadmill exercise, increasing load experimental animal model(grade 0., increasing exercise intensity to 21 m/min x 45 min, exercise intensity is equivalent to 75 ℅ VO2max), continuous training for five days A week, A total of 8 weeks.Groups of rats in 24 h after the last training session anesthesia, broken neck death, materials.Will be fixed on the operating table of rats after death, cut the skin of lower limb, careful handling mouse hair, avoid sticking on the muscle, quickly remove the quadriceps.In the cold saline to remove fat and other connective tissue, filter paper blot, put in liquid nitrogen freezing,- 80 ℃ low temperature preservation.Using colorimetric method were determined mitochondrial atpase activity and mitochondrial respiratory chain complexes III, IV activity, mitochondria, inhibits PTP opening degree, electron microscope mitochondria structure change.The results of the study:(1) skeletal muscle mitochondria inhibits PTP opening degree: quiet control group with incremental load aerobics group, the incremental load aerobics + curcumin is all have significant difference(P < 0.05), quiet curcumin group compared with incremental load aerobic exercise group had significant difference(P < 0.05), increasing load increasing aerobic exercise group and load the difference between aerobic exercise and curcumin group is not obvious, but presents the tendency of decrease.(2) The skeletal muscles mitochondrial ATP enzyme activity: quiet quiet control group with curcumin, incremental load aerobics group, incremental load aerobics and curcumin is had extremely significant difference(P < 0.01), increasing load increasing aerobic exercise group and load between aerobic exercise and curcumin groups have significant difference(P < 0.05).(3) The skeletal muscles mitochondrial CIII activity: aerobic exercise group quiet control group with increasing load, incremental load aerobics and curcumin is had extremely significant difference(P < 0.01), quiet curcumin group compared with incremental load aerobic exercise group was extremely significant difference(P < 0.01), quiet curcumin group compared with incremental load aerobics + curcumin groups was very significant difference(P < 0.01);Increasing load aerobics group compared with incremental load aerobics and curcumin groups had significant difference(P < 0.01).(4) The skeletal muscles mitochondrial CIV activity: quiet control group compared with quiet curcumin groups had significant difference(P < 0.05), the quiet control group compared with incremental load aerobic exercise group had extremely significant difference(P < 0.01), the quiet control group compared with incremental load aerobics and curcumin groups was very significant difference(P < 0.01), the static curcumin incremental load aerobics group comparison has the extremely significant difference(P < 0.01), quiet curcumin group compared with incremental load aerobics and curcumin groups was very significant difference(P < 0.01), increasing load aerobics group compared with incremental load aerobics and curcumin groups have significant difference(P < 0.01).Research conclusions:(1)curcumin aerobic exercise combined with incremental load to middle-aged and old rats quadriceps showed a trend of reduce mitochondrial inhibits PTP opening degree.(2)Curcumin aerobic exercise combined with incremental load to middle-aged and old rats quadriceps mitochondrial ATP enzyme activity increased.(3)Curcumin combined with incremental load aerobics in middle-aged quadriceps CⅢ activity of mitochondria in rats(4)Curcumin combined with incremental load aerobics older quadriceps CⅣ activity of mitochondria in rats.
