With the rapid development of the food industry in recent years,food additives have become an indispensable element for the food production.It will be helpful to the diversification of food and improving the quality of food using properly food additives.However,long-term consumption of food with additives used illegally or excessively,it will lead to dangers such as carcinogenicity and genotoxicity.Currently,conventional analytic methods have not meet the needs of increasingly heavy workload as a widespread use of multiple food additives.Thus,it is urgent to develop high-efficient analytic methods for a rapid detection of multiple additives simultaneously.Compared with High Performance Liquid Chromatography(HPLC),Ultra-high Performance Liquid Chromatography(UPLC)has the advantages of shorter analysis time,higher separation efficiency and less consumption of solvent,and it is more suitable for the simultaneous detection of multiple targets.In this paper,based on the advanced technology of UPLC,methods for the multi-detection of food additives,such as synthetic colors,antioxidants,preservatives and sweeteners,were established by simplifying pretreatment operations of sample and optimizing separation conditions of chromatography.Results showed that the methods were simple,fast,highly efficient.The main results and conclusions are as follows.1)The method for detecting 10 kinds of synthetic colors based on UPLC was established.Sample was firstly extracted with mixed solution of Ammonia and ethanol,then was purified by strata X-AW SPE columns.Using Agilent ZORBAX Eclipse Plus C18(2.1 x 50 mm,1.8 m)column as separation column and methanol-0.02 mol/L ammonium acetate solution as mobile phase,10 kinds of synthetic colors were separated by gradient elution and quantified by external standard method.Results showed that the retention time of compounds were all less than 4 min.Each compound had a good linearity in the range of 1~50 ?g/m L,R~2?0.998.Detection limits were 0.004~0.022 mg/kg.Adding 3 different levels of 4,8,16 mg/kg,the average recovery rate is 83.75~111.25%,and the RSD is 1.24~4.18%.This method with high flux and less interference,is accurate and reliable.2)The method for detecting 4 kinds of phenolic antioxidants in oil samples based on UPLC was established.The sample was dissolved in hexane and re-extracted with acetonitrile many times.Using ZORBAX Eclipse XDB-C18(2.1×50 mm,1.8-Micron)column and the determination of methanol-1.0% acetic acid aqueous solution to carry out gradient elution,antioxidants were separated and quantified by external standard method.Results showed that the retention time of antioxidants were all less than 6 min.Each compound had a good linearity in the range of 0.5~25 ?g/m L,R~2?0.998.Detection limits were 0.05~0.16 mg/kg.Adding 3 different levels of 50,100,150 mg/kg,the average recovery rate is 88.18~100.25%,and the RSD is 1.23 ~ 4.28%.This method has advantages of convenient operations,widespread feasibility and high efficiency,and the detecting results was reliable and sensitive.3)The method for detecting 7 kinds of preservatives and 2 sweeteners in food based on UPLC was established.Sample extraction was separated by the column of Waters ACQUITY UPLC BEH C18(2.1×100 mm,1.7 ?m)and gradient elution method with methanol-20 mmol/L ammonium acetate solution as mobile phase,finally quantified by the external standard method.Results showed that the retention time of compounds were all less than 13 min.Each compound had a good linearity in the range of 5~50 ?g/m L,R~2?0.998.Detection limits were 0.015~0.052 mg/kg.Adding 3 different levels of 100,200,300 mg/kg,the average recovery rate is 83.10~105.21%,and the RSD is 1.98~6.08%.This method is saving time and effort,with the strong ability of anti-interference,which can meet the demand of batch testing work in the laboratory for it is accurate and reliable.
