Ganoderma philppiii belongs to the genus Ganoderma of the family Ganodermataceae,which mainly distrited in tropical regions,such as Hainan in China,Malaysia,Philippine,etc.It usually grows on the genus rubber(Hevea)and palm plants.As one of most important Ganoderma resources in Hainan,it has been widely used in folk.However,study on chemical constituents and biological activities of Ganoderma philppiii have not been reported.Coprinus setulosus is a common mushroom.However,the chemical constituents and biological activities from the fruiting bodies of genus Psathyrellaceae were not barely reported until now.In order to better exploitating Ganoderma philppiii resources,as well as providing scientific basis for the innovative use of Coprinus resources in Hainan Island.We chose these two fungus as research object to study their chemical constituents and biological activities.Twenty-three compounds were isolated from the EtOH fraction of the fruiting bodies of Ganoderma philppiii by various chromatographic methods(silica gel column chromatography,RP C-8 column chromatography,Sephadex LH-20 column chromatography,preparative-HPLC,et al.).Among them,there are three new compounds and their structures were identified as ganoderin B?26-nor-11,22-dioxo-5?-lanost-8--en-3?,7?,15?-triol?3?,9?,14?-trihydroxy-(22E,24R)-ergost-22-en-7-one on the basis of modern spectrometric approaches,including 1D NMR,2D NMR,UV,IR,EI-MS,etc.Eleven compounds were isolated from the ethanol extract of the fruiting bodies of Coprinus setulosus by various modern chromatographic techniques.A new compound was isolated,and its structure was identified by physicochemical and NMR analysis as ergosta--4,6,8(9),22E-tetraen-11?-ol.All compounds were found to be isolated from the fruiting bodies of Ganoderma philppiii and Coprinus setulosus for first time.The cytotoxic activities of 26-nor-11,22-dioxo-5a-lanost-8-en-3?,7?,15?-triol was measured by MTT assay,which showed no obviously inhibitory activities against human chromic myeloid leukemia(K562),human liver cancer(SMMC-7221)and human lung cancer(A549).The bioassay of inhibitory activity against acetylcholinesterase(AChE)of four new isolates were tested,and ganoderin B and ergosta-4,6,8-(9),22E-tetraen-11?-ol showed no obviously inhibitory activity against AChE;26-nor-11,22-dioxo-5?-lanost-8--en-3?,7?,15?-triol exhibited lowly inhibitory activity,while 3?,9?,14?-trihydroxy-(22E,24R)-ergost-22-en-7-one presented definite inhibitory activity.
